The comedy centers on Glen "Fitz" Fitzgerald (James Pumphrey), a young man, marijuana dealer, and former rock band drummer living in Los Angeles, California. His best friends dropped out of the band, Torigl, three months prior, causing the group to part and him to fall back on selling weed in his garage. He dates Monica (Abby Elliott), who works at a news company, and hangs out with his rebellious teen neighbor, Jimmy (Dylan O'Brien), who skips school. Fitz also believes in the "triangle theory", where everything in the world can connect in group of threes (a joke ran throughout the film). One day, after Fitz is nearly arrested in a weed bust of one of his clients, he leaves his girlfriend behind and flees to temporarily stay in Oakland, California to avoid arrest. Jimmy convinces him to join his trip so he can visit his mother there, as his father James Malone Sr (Rob Riggle) plans to send him to military school for missing and failing classes.

Ted Wiggins is an idealistic 12-year-old boy [citation needed], who lives in "Thneedville", a walled city that, aside from the human citizens, is completely artificial; everything is made of plastic, metal, or synthetics. Ted has a crush on local environmentalist Audrey, who wants to see a "real tree" more than anything in the world, and decides to find one in order to impress her. His energetic Grammy Norma secretly tells Ted the legend of the Once-ler, who will tell anyone about trees if you brought him fifteen cents, a nail, and a shell of a great-great-great grandfather snail. When Ted leaves Thneedville in search of the Once-ler, he discovers that the outside world is a contaminated, empty wasteland. Once the boy finds him, the Once-ler agrees to tell Ted about the trees on the condition that he listens to the story over multiple visits. Ted agrees, but on his way home, he encounters the mayor of Thneedville, Aloysius O'Hare, who is also the proprietor of a company that sells bottled oxygen to Thneedville residents. O'Hare explains to Ted that because trees produce oxygen free of charge, he considers it a threat to his business whenever he hears people talking about them. After revealing that he has "security camera eyes" all over the city, O'Hare pressures Ted to stay in town. However, Ted continues to sneak out of O'Hare's sight (with his grandmother's encouragement) and learns more of the trees' history.

Two years after the events in Bangkok, Leslie Chow (Ken Jeong) escapes from a maximum security prison, using a riot as cover. On the way to Arizona, Phil's minivan is rammed off the road by a rental truck and the group is taken hostage. They are later confronted by mob boss Marshall (John Goodman) and "Black Doug" (Mike Epps), his head enforcer.

A low-level marijuana dealer named David Clark (Jason Sudeikis) who lives in Denver is robbed of his money and stash, some of which he owes to his wealthy drug lord supplier Brad Gurdlinger (Ed Helms). After David is brought to him, Gurdlinger forces David to smuggle marijuana from Mexico as a solution to clear his debt. Realizing that one man attempting to get through customs is too suspicious, he hires a middle-aged stripper neighbor stage-named Rose (Jennifer Aniston), a runaway and petty thief named Casey (Emma Roberts), and his 18-year-old neighbor Kenny (Will Poulter), to pose as a bogus family called the "Millers". When the Millers reach the compound, they find out that the amount of marijuana they were picking up is not "a smidge" as Gurdlinger had informed them, but actually two tons. Because of the extra load of the marijuana on the RV, one of the radiator hoses breaks while going up a steep incline. A family they had encountered at the border, the Fitzgeralds, consisting of Don (Nick Offerman), Edith (Kathryn Hahn), and Melissa (Molly Quinn), catch up to them and tow the Millers' RV to a repair shop. On the trip to the shop, David learns that Don Fitzgerald is a DEA agent after finding his badge and gun in the glove compartment.
