Architect Leo Waters' life is in trouble and in order to have some sense of control, he attempts to lord over the other members of his family. His career appears to be going nowhere; his wife Julia (Isabella Rossellini) is a bored housewife who spends her time tending to the luxurious modern house he has designed for them, their son Martin (Sebastian Stan} drops out of college and has no interest in taking up his father's dream of also becoming an architect, and their daughter Christina (Hayden Panettiere) has entered her mid-teens and her father has started staring at her maturing body in an unfatherly way.

The film is set in a dark and decadent area of São Paulo, Brazil, where the exiled Americans Sinatra and his son Paul own a brothel. Paul is a compulsive gambler addicted to cocaine. Sinatra is married to a former prostitute named Angie, with whom he has a son. A Russian client is killed by his wife in their establishment, leaving behind a suitcase filled with drugs. On the night that they have scheduled a negotiation to sell the contents of the suitcase to African buyers, their go-between dies while having sex with a transvestite named Nazda. In desperation, Sinatra makes a deal with the Nigerian dishwasher of the brothel, Wemba. Wemba is to travel to the harbor of Santos, taking the place of the go-between, and make the sale to the drug dealers. In return Wemba would receive a large amount of money. Wemba accepts but while returning to his car in the harbor, he is attacked by two small-time thieves and is knocked unconscious. His lack of contact with Sinatra and Paul starts a chain-reaction of misunderstandings that lead to a tragic end.

In late 19th century Cartagena, a river port in Colombia, Florentino Ariza falls in love at first sight with Fermina Daza. They secretly correspond and she eventually agrees to marry but her father discovers their relationship and sends her to distant relatives (mainly her grandmother and niece). When she returns some years later, Fermina agrees to marry Dr. Juvenal Urbino, her father's choice. Their fifty-year marriage is outwardly loving, but inwardly marred by darker emotions. Fermina's marriage devastates Florentino, who vows to remain a virgin, but his self-denial is thwarted by a tryst. To help him get over Fermina, his mother throws a willing widow into his bed and he discovers that sex is a very good pain reliever with which he replaces the opium that he had habitually smoked. He begins to record and describe each of his sexual encounters, beginning with the widow, and eventually compiling over 600 entries. A lowly clerk, he plods resolutely over many years to approach the wealth and social standing of Dr. Urbino. When the now-elderly doctor dies suddenly, Florentino immediately and impertinently renews courting Fermina.

The film spans several decades in the unconventional life of feisty nonagenarian Hagar Shipley, who sets off on a journey to reconcile herself with her past when she discovers her son Marvin and daughter-in-law Doris are moving her into a nursing home. Mustafa (Sayed Badreya) is a widowed Egyptian immigrant and the owner of Habibe's Café, a popular hangout in Los Angeles for those with Middle Eastern backgrounds. He is devoted to providing his son, Richard Chagoury, with a moral upbringing despite the pressures of contemporary American urban life. He also finds himself cast in the role of protector to his unwed sister Salwah (Sarah Shahi), for whom, by family and tribal custom, he is responsible for finding a traditional suitor. But his respect for tradition comes up against his own aspirations to adapt to the American Dream when he decides to open a new restaurant with a Jewish partner – his friend Sam (Tony Shalhoub). This unholy alliance is unpopular amongst the habitués of his café and the insular Arab community in which Mustafa resides. It is one of several personal points of tension that gradually build against the backdrop of larger, national events affecting the Arab-American community and lead to the explosive denouement of the story.
