Dr. Peter Carey (James Coburn) is a pathologist who moves to Boston, where he starts working in a hospital. He soon meets Georgia Hightower (Jennifer O'Neill), with whom he falls in love. Karen Randall, daughter of the hospital's Chief Doctor, becomes pregnant and is brought to the emergency department after an illegal abortion. She dies there, and Dr. David Tao (James Hong), a brilliant surgeon and friend of Carey, is arrested and accused of being responsible for the illegal abortion. Carey does not believe his friend to be guilty and starts investigating on his own, despite strong opposition by the police and the doctors around the hospital's chief.

Philippe Douvier (Robert Webber), a major businessman and secretly the head of the French Connection, is suspected by his New York Mafia drug trading partners of weak leadership and improperly conducting his criminal affairs. To demonstrate otherwise, Douvier's aide Guy Algo (Tony Beckley) suggests a show of force with the murder of Chief Inspector Jacques Clouseau (Peter Sellers). Unfortunately for Douvier, his first attempt at bombing him fails; and the subsequent attempt by Chinese martial artist 'Mr. Chong' (an uncredited appearance by the founder of American Kenpo, Ed Parker) is thwarted when Clouseau successfully fights him off, believing him to be his (Clouseau's) valet Cato (Burt Kwouk) who has orders to keep his employer alert with random attacks. That night, Douvier poses as an informant to lure Clouseau into a trap, but the Chief Inspector's car and clothes are stolen by transvestite criminal Claude Russo (Sue Lloyd), who is killed by Douvier's men. Subsequently Douvier and the French public believe Clouseau is dead and, as a result of this assumption, Clouseau's former boss, ex-Commissioner Charles Dreyfus (Herbert Lom), is restored to sanity and is released from the lunatic asylum to perform the investigation.

In the fictional country of Lugash, a mysterious thief seizes the Pink Panther diamond and leaves a white glove marked with a gold-tinted "P". With its national treasure once again missing, the Shah of Lugash requests the assistance of Inspector Clouseau (Peter Sellers) of the Sûreté, as Clouseau had recovered the diamond the last time it was stolen. Clouseau has been temporarily demoted to beat cop by his boss, Chief Inspector Charles Dreyfus (Herbert Lom), who despises him to the point of obsession. But after the request, the French government forces Dreyfus to reinstate him. Clouseau joyously receives the news after fending off a surprise attack from his servant Cato (Burt Kwouk), who had been ordered to keep the Inspector on his toes, and duly goes to Lugash.

An attractive British Home Office assistant named Judith Farrow (Julie Andrews) is on vacation on the Caribbean island of Barbados after ending a failed love affair with married group captain Richard Paterson (David Baron), an important British minister. She meets Feodor Sverdlov (Omar Sharif), a Soviet military attaché who is also on vacation staying in an adjacent bungalow. The two spend time together exploring the island, visiting museums, and going out to dinner. When British intelligence learns that Sverdlov is spending time with the mistress of a British minister, they begin monitoring their actions. Judith and Sverdlov share details about their private lives—about her husband who died in a car crash, her recent unhappy affair, his unhappy marriage, and his disillusion with the Soviet Union. During one of their outings, Judith becomes fascinated by the story of a slave who was hanged from a tamarind tree and how that tree has since borne seeds in the shape of a human head. When the famous Pink Panther diamond is stolen again from Lugash, Chief Inspector Clouseau (Peter Sellers) is called on the case despite protests by Chief Inspector Dreyfus (Herbert Lom). While on the case, Clouseau is pursued by the Mafia. Clouseau first goes to London to interrogate Sir Charles Lytton (having forgotten that he lives in the South of France). Traveling to the airport, he accidentally blows up his car, but mistakenly believes it an assassination attempt, and disguises himself in a heavy cast on the flight, which causes complications in the air and on land and leads to an awkward introduction to the Scotland Yard detectives at Heathrow. Meanwhile, Dreyfus learns from Scotland Yard that Libyan terrorists have marked Clouseau for assassination; but permits him to continue.

As a child, Princess Dala receives a gift from her father, the Shah of Lugash--"the Pink Panther," the largest diamond in the world. This huge pink gem has an unusual flaw: looking deeply into the stone, one perceives a tiny discoloration resembling a leaping panther. A promiscuous and rather misogynistic man, Steve (Perry King) is murdered by one of his three ex-lovers, and after death, the powers that be cannot decide whether to send him to heaven or to hell—his life is full of good deeds, but he's "been a shit" to women, and that behavior is keeping him from going to heaven. The powers that be decide to give him a test; he is reincarnated and given a limited amount of time to have at least one woman be in love with him. To make the test more difficult, he is reincarnated as a beautiful woman (Ellen Barkin) named Amanda. After the change, Amanda/Steve encounters other people, including Steve's friend Walter, who Amanda convinces of the truth of her identity, and lesbian perfume magnate Sheila. Both become attracted to Amanda, but Amanda rebuffs Sheila's advances, even knowing that she is giving up an opportunity for a woman to love her, because Steve is homophobic and could not abide having gay sex, even though he had sex exclusively with women, as Steve, in the past. Later, Amanda and Walter get drunk together and end up having sex. In the morning, Amanda claims no memory of the sex and accuses Walter of raping her. Walter acts surprised and insists that Amanda was an enthusiastic participant. Amanda rejects Walter, but she learns shortly thereafter that she has become pregnant from the encounter. At the childbirth, the baby girl gazes upon her mother with love, and then Amanda dies, having earned her place in heaven.
