At the start of the film, the US Government has ordered a branch of the US Military to discontinue tests concerning "the C.H.U.D. project", which is built around the idea that enzymes taken from the sewer dwelling creatures from C.H.U.D. can make hyper-effective killing machines in the army. For reasons that are unclear even to those who watch the film, the last specimen of the experiment (BUD the C.H.U.D.) is hidden away in a Centre for Disease Control in a small American town, where a trio of bumbling teenagers steal and accidentally reawaken him. Bud escapes and begins to forge an army of C.

To ensure the snakes attack the passengers without being provoked, he has one of his henchmen disguised as an airport ground employee spray the passengers' leis with a special pheromone which makes the snakes highly aggressive. The crate opens midway through the flight and the snakes make their way through the cabin. A couple having sex in the bathroom, and a man using the bathroom are the first killed. The plane's captain, Sam McKeon, investigates and fixes an electrical short, but is killed by the viper that caused it. Co-pilot Rick, unaware of the snake, believes Sam has suffered a heart attack and continues toward LAX.

Entomologist Dr. James Atherton (Julian Sands) searches the Amazon rainforest with the hope of discovering new species of insects and arachnids. Descending into an enormous sinkhole, his team sets up collectors on the ground below a large tree, then blow smoke up into the canopy of the tree. A large number of bugs fall into the collector, including a very aggressive new species of spider. The spider is captured and chloroformed for research; and is later revealed to be lacking sex organs, thus making it a drone, or soldier, highly unusual in an arachnid species. A nature photographer, Jerry Manley (Mark L. Taylor), who has been suffering a fever while traveling with the team, unknowingly has a fertile (non-drone) male spider of the same species jump into his backpack, that night sneaking into his sleeping bag and biting him. Manley has a massive seizure from the venom and dies. The remainder of the scientists takes his body back to the United States, blaming Manley's death on the preexisting fever. The spider crawls into the box and is sealed in with the corpse.
