Romeo Rajkumar (Shahid Kapoor) is an aimless youth who arrives in small town of Dhartipur been ruled by two drug barons named Shivraj Gurjar (Sonu Sood) and his arch enemy Manik Parmar (Ashish Vidyarthi), controlled by a Mafia don named Ajit Taaka (Srihari), who operates in Hong Kong. Rajkumar now works for Shivraj. But his life changes forever when he claps eyes on the beautiful, educated Chanda (Sonakshi Sinha). What he does not know is, that Chanda is an orphan who is being raised by Manik Parmar. After some persistent wooing, Chanda falls for Rajkumar's charms. Just when their love was about to flourish, Shivraj and Parmar compromise and Parmar decides to get Chanda married to Shivraj, against Chanda's wish. Angered Rajkumar beats Shivraj's men and challenges him, saying that he will marry Chanda in front of Shivraj. Furious, Shivraj plans to get Rajkumar killed in an encounter by his pet police officer. But in return, gets a shocking surprise when it is revealed that Rajkumar actually works for Don Ajit Takka and was sent to Dhartipur only to take over the drug cartel to which both Shivraj and Parmar owe allegiance. Rajkumar captures Shivraj's truck full of drugs and drives it to Takka, revealing he himself was a big gangster all along. After giving Shivraj's stolen truck to Takka, he discovers that the truck was empty. In a twist of events, it turns out that Shivraj and Takka had joined hands, and purposely made Rajkumar steal the empty truck. Shivraj and Takka brutally kill Rajkumar and bury him. Shivraj starts his wedding celebrations with Chanda, only to be interrupted by the still alive Rajkumar, who is shown to have been rescued by one of Shivraj's henchmen after he was buried. Shivraj's men back off, because they are now friends with Rajkumar, hence leading to a one on one fight between the two. Finally, Rajkumar injures Shivraj with a power punch to the neck and later kills Don Takka with a ceramic tile. The film ends with all the men beating Parmar, while a severely injured Rajkumar and Chanda walk away hand in hand happily.

Siddharth (Farhan Akhtar) and Trisha (Vidya Balan) are a happily married couple until Trisha gets pregnant and things start getting complicated. Sid is not ready for a baby as he is just a struggling music composer, yet lies to Trisha. On his friends' advice, Sid starts reading pregnancy books and following Trisha's daily routine by putting a balloon in his shirt. Finally, Trisha gives birth to a baby girl and starts devoting all her time to the baby. Trisha's mother (Rati Agnihotri) feels that Sid is a careless person and can not take this responsibility, at which Sid makes a desperate attempt to behave like a family man but fails, as on one occasion he takes his baby for a walk but forgets her in the market. Frustrated, Sid ask for help from Trisha's brother-in-law Ranveer (Ram Kapoor), who is a typical family man. Ranveer reveals to him that when he gets frustrated, he makes a business trip excuse and stays in a hotel room to enjoy his life as a bachelor. Sid takes on his idea and starts staying in a hotel, with the excuse of working overnight in studio. But soon he has to cut down his idea, due to an added expense at home as Trisha decides to hire an expensive governess (Ila Arun). Sid, then, without telling Trisha, goes for a cheaper solution by shifting to a shared PG along with Manav (Vir Das), a dope-head bachelor. Sid takes Manav's advice and starts living a carefree life.

Prabhu Sivaanu (Siddharth) and his friends discuss the route from Bangalore to Chennai and plan to kidnap Rithika (Ashrita Shetty). Meanwhile her dad appoints encounter specialist Manoj Menon (Kay Kay Menon) who questions Rithika's male friend Deepak (Deepak) about Prabhu. The guy claims that Prabhu is a drug addict and was always trying to woo Rithika, who started to avoid him because of Prabhu. After that the plot continues when Manoj tries to track Prabhu and his friends. In the attempt, they track down a message from another friend Umesh asking Prabhu and friends to come to Bangarpet bus stand only to find Manoj and his team arresting the guy for more information. Manoj goes to a railway station where the train in which Prabhu and Rithika are travelling to Chennai. He finds Rithika and forcefully drags her into his jeep. As the flashback winds again (this time by Rithika), she fell for Prabhu after seeing him on many instances showing his presence of mind. As the flashback ends, Manoj finds Prabhu on the highway where he doesn't stop until he runs over pieces of glass put down by Prabhu. They get into a brawl where Prabhu loses and Manoj continues in the highway but has to stop in a railway signal. Prabhu takes his chance and threatens Manoj keeping his throat under a blade and taking Rithika with him. After a cat and mouse chase, Manoj calls Prabhu saying that his friend needs immediate treatment after an accident. He goes in search of Manoj and involves in a fight with him again. Now, after the stroke of midnight Rithika turns 18 giving her the right for choosing her partner. After that Manoj leaves the Highway as he couldn't do anymore according to law.
