The Joker and the Penguin break out of Arkham Asylum and race each other to find a stash of stolen money hidden within a crypt in Gotham Cemetery after an inmate, who had hidden it, told them about it. The Joker is quickly intercepted by the Batman and is electrocuted when he falls into a river by his super-charged joy buzzers while the Penguin takes the opportunity to enter the cemetery himself. While searching through a crypt, the Penguin accidentally cuts his hand while using his umbrella-sword to open a coffin he hopes contains the money, finding a body instead. Blood from his hand drips on the corpse's heart, which brings it to life; it is none other than the vampire lord Count Dracula, his body having been moved from Transylvania to Gotham City after his "death." After attacking and turning a watchman at the cemetery into a vampire, Dracula hypnotizes the Penguin into becoming his non-vampiric servant so that he may lead him through Gotham to feed and guard his coffin during the day while he regains his strength.

The piece begins with the view of the Wayne Enterprises building to which Bruce Wayne is gazing out the window during a party. He's pulled to the dance floor by three lovely young ladies. Each has their turn before Alfred rescues Bruce, who dives into a nearby elevator. When he reaches his office, he sees none other than Catwoman at the safe, looting it. Catwoman pins Bruce to the wall while she finishes robbing the safe, and makes her escape. Bruce breaks free, and heads after her as Batman.

While no cohesive plot is apparent from the vignette style of the trailer, it can be pieced together that Batman has been murdered, and his killer remains at large. Dick Grayson, long since retired from his superhero days and married to Barbara Gordon with a child, decides to resume his crime-fighting days as Robin; Remarkably, Grayson does not take up the Nightwing identity. The filmmakers said they chose this because many people outside the comic book community are unfamiliar with Nightwing and they wanted to appeal to a wider audience.

Robin is waiting for a big date in a restaurant. Unfortunately, Batman (who Robin desperately tries to convince not to show up, as he'll ruin everything) shows up. Kate, his date, arrives at the restaurant shortly after Batman, and the Dark Knight becomes incredibly obnoxious toward her, dominating the conversation and flirting. The Justice League, with assistance from Cyborg, stop the Royal Flush Gang as they attempt to rob a diamond vault using complex technology that allows them to pass through solid objects. Wonder Woman uses her lasso of truth on the leader to find out who gave them the technology. The leader, despite being under the influence of the lasso, says that he doesn't know who gave them the technology. Martian Manhunter uses his mental powers to scan the minds of each member of the gang, none of them know the origin of the device. Meanwhile, Vandal Savage plots to exterminate the greater part of the human population and start a new civilization, having discreetly given the technology to the Gang in order to test it. To ensure that the Justice League is unable to stop him, Savage hires Mirror Master, who hacks into the Batcomputer and steals contingency plans devised by Batman to incapacitate his League teammates should they ever go rogue. When Mirror Master asks if he wants him to find Batman's true identity, Savage declines stating that he already knows the true identity of Batman. Savage assembles Mirror Master and a group of other supervillains (consisting of Bane, Cheetah, Ma'alefa'ak, Star Sapphire, and Metallo) with personal vendettas against the heroes and pays them to simultaneously attack the members of the League using the stolen plans, albeit altered to be lethal. When the supervillains agree with Savage's plan, he welcomes them to the Legion of Doom.

Batwoman employs the services of several young female agents known as "Batgirls", in her pursuit of justice. Her archenemy is a masked villain named "Rat Fink". Added to the mix is the President and Vice-President of the "Ayjax" Development Corporation. The company, using plutonium as its fuel source, has created a powerful listening device called "The Atomic Hearing Aid", which allows for limitless eavesdropping. The company has been ordered to destroy the hearing aid by the US Government when the government turned down the offer to buy the listening device (due to its unstable power supply) but the President of the company has refused to do so and now is being confronted by Rat Fink about selling him the device.
