In a dystopian near-future, Detroit, which is now bankrupt and overrun with crime, gives Omni Consumer Products (OCP) control of its struggling police force. The company plans to replace the poor, run-down sections of Old Detroit with the high-end "Delta City," but must first address the city's high crime rate. As an alternative to existing law enforcement, OCP senior president Dick Jones (Ronny Cox) offers the prototype ED-209 enforcement droid, but it accidentally kills a board member during a demonstration. The OCP chairman, nicknamed "The Old Man" (Dan O'Herlihy), decides instead to back Jones' young rival, Bob Morton (Miguel Ferrer), and his experimental cyborg police officer program, "RoboCop.

In 2010, United Nations negotiator Edward Douglas (David Thewlis) survives a plane crash in the Java Sea and is eventually rescued by a passing boat. Aboard, a man called Montgomery (Val Kilmer) tends to him, and after telling him the boat has no radio, he promises Douglas the captain will take him to Timor after dropping him off. However, when they arrive at Montgomery's destination referred to as "Moreau's Island", he instead advises Douglas to stay with him, ostensibly so he can use the radio on the island.

Chev Chelios (Jason Statham) is a Chinese-based British hitman, working for a crime syndicate headed by Uglyman (Carlos Sanz). Carlito betrays Chelios by making it appear he assassinated Don Kim on his own, without his authorization. Ricky Verona (Jose Pablo Cantillo), an ambitious small-time criminal uses the opportunity to conspire with Carlito against Chelios; Verona will kill Chelios so the Triads do not retaliate, and then take Chelios's place as Carlito's new hired gun. While Chelios sleeps in his apartment, Verona, his brother Alex (Jay Xcala), and several henchmen break in and inject Chelios with a Chinese synthetic drug which inhibits the flow of adrenaline, slowing the heart and eventually killing the victim. Chelios wakes to find a recording left by Verona showing what he has done.

Shekhar Subramanium (Khan), a game designer who works for the London-based Barron Industries, has delivered a number of commercial failures; an irate Barron (Tahil) gives him his last chance to develop a successful game. His colleague, computer programmer Jenny (Goswami), uses Shekhar's face as a model for that of the game's protagonist G.One (Technically Good One and in Hindi Jeevan, which means life), while the shape-shifting antagonist Ra.One (Technically Random Access Version One and in Hindi Ravan, a mythical demon) is made faceless. Another colleague, Akashi (Wu), implements the characters' movements. The game, named Ra.One, contains three levels, the final level being the only one in which either character can be killed. Each character possesses a special device – the H.A.R.T (Hertz Amplifying Resonance Transmitter) which gives them their powers. Upon reaching the last level, the characters gain a gun with one bullet; the other character can be killed by this bullet but only if his H.A.R.T is attached.

However, since its inception the MCP has become power-hungry, illegally appropriating business and even government programs and absorbing them to increase its own capacities; it informs Dillinger of its plans to subjugate The Pentagon and the Kremlin, and expresses interest in China with its request for Chinese-translation programs, blackmailing Dillinger into compliance with records of his theft of the games.

This caused Barnes to become a convicted killer and serve 17 years to life.
