Fred Wilson (Charles Grodin), an executive of the Petrox Oil Company, forms an expedition based on infrared imagery which reveals a previously undiscovered Indian Ocean island hidden by a permanent cloud bank. Wilson believes the island has a huge deposit of oil. Jack Prescott (Jeff Bridges), a primate paleontologist who wants to see the island for himself, stows away on the expedition's vessel. Prescott reveals himself when he warns the crew the cloud bank may be caused by an unknown beast. Wilson orders Prescott locked up, claiming that he is really a spy from a rival oil company. While escorted to lock-up, Prescott spots a life raft which carries the beautiful and unconscious Dwan (Jessica Lange). Wilson conducts a thorough background check on Prescott and realizes he is telling the truth. He appoints Prescott the expedition's official photographer and requests that he be present when Dwan revives because of his medical background. Upon waking, Dwan says she is an aspiring actress who was aboard a director's yacht which suddenly exploded.

Mr. Tako, head of Pacific Pharmaceuticals, is frustrated with the television shows his company is sponsoring and wants something to boost his ratings. When a doctor tells Tako about a giant monster he discovered on the small Faro Island, Tako believes that it would be a brilliant idea to use the monster to gain publicity. Tako immediately sends two men, Sakurai and Kinsaburo, to find and bring back the monster. Meanwhile, the American submarine Seahawk gets caught in an iceberg. The iceberg collapses, unleashing Godzilla (who, in the Japanese version, had been trapped within since 1955), who then destroys the submarine and a nearby military base.

In New York Harbor, Carl Denham, famous for making wildlife films in remote and exotic locations, charters Captain Englehorn's ship Venture for his new project, but he is unable to secure an actress for a female role he reluctantly added. Denham searches the streets of New York for a suitable woman. He meets penniless Ann Darrow and convinces her to join him for the adventure of a lifetime. The Venture quickly gets underway. The surly first mate, Jack Driscoll, gradually falls in love with Ann. After weeks of secrecy, Denham finally tells Englehorn and Driscoll that their destination is Skull Island, an uncharted island shown on a map in Denham's possession. Denham speaks of something monstrous there, a legendary entity known only as "Kong".

King Kong, after being shot down from the World Trade Center, is kept alive in a coma for about 10 years at the Atlanta Institute, under the care of surgeon Dr. Amy Franklin (Linda Hamilton). In order to save Kong's life, Dr. Franklin must perform a heart transplant and give Kong a computer-monitored artificial heart. However, he lost so much blood that a transfusion is badly needed, and to complicate matters, Franklin says there no species of ape or other animal whose blood type matches Kong's. Enter adventurer Hank "Mitch" Mitchell (Brian Kerwin), who captures a giant female gorilla, who is dubbed "Lady Kong"; in Borneo (Mitchell theorizes that Borneo and the island from the first movie were once part of the same landmass), bringing her to the Institute so her blood can be used for Kong's operation. The transfusion and the heart transplant are a success, but Kong escapes along with the female.

An evil genius named Dr. Who creates Mechani-Kong, a robotic version of King Kong, to dig for a highly radioactive Element X, found only at the North Pole. Mechani-Kong enters an ice cave and begins to dig into a glacier, but the radiation destroys its brain circuits and the robot shuts down. Who then sets his sights on getting the real Kong to finish the job. Who is taken to task by a beautiful female overseer, Madame Piranha. Her country's government is financing the doctor's schemes, and she frequently berates him for his failure to get results.

The story picks up about a month after the dramatic finale of the previous film and follows the further adventures of filmmaker Carl Denham, now implicated in numerous lawsuits following the destruction wrought by Kong. Denham leaves New York with the captain of the Venture, Captain Englehorn, who is certain it is just a matter of time before he is similarly served. Their efforts to make money shipping cargo around the Orient are less than successful. In the Dutch port of Dakang, Denham is amused to see there's a "show" being presented, so he and Englehorn attend. It turns out to be a series of performing monkeys, capped by a song ("Runaway Blues") sung by a young woman named Hilda.

A 36-foot-gorilla escapes from an oil tanker off the coast of Korea. After battling with a giant Great White Shark, the ape reaches land and destroys several buildings and attacking a giant Python (the snake's origins are never revealed) before finding and kidnapping an American actress named Marilyn (Joanna Kerns). The military eventually corners and kills the ape, prompting one observer to comment, "He was just too big for a small world like ours!"

One night Chinami (Reiko Mishima), the daughter of Hyoue Toba (Reizaburo Ichikawa), is mysteriously kidnapped. Hyoue offers a large reward for his daughters rescue. Yuzuru Kawasaki (Noboru Takashima) and various other men employed by Hyoue set about searching for Chinami. However one of Hyoue's men, Magonojyō Gō (Eizaburo Matsumoto), doesn't partake in the search. Why? Because he was the one who had Chinami kidnapped. Magonojyō's father Senbei has a pet trained ape named "King Kong" (Ryūnosuke Kabayama) and Magonojyō used this creature to perform the kidnapping.

The film begins with Ann Darrow, a down-on-her-luck actress looking for work, meeting film director Carl Denham, who offers her a job in a new movie. They board the Venture to leave for the film shoot. The monkey that lives on board causes trouble throughout the trip. They arrive on the island and the natives, who are friendly to the crew, ask that the crew leave Ann so they can sacrifice her. Ann is then sacrificed to the giant ape King Kong who makes off with her into the jungle. Together they fight a dinosaur and are off the island in five minutes.

Santa (Isamu Yamaguchi) and his friend Koichi (Nagamasa Yamada) are jobless vagabonds. They make their living by picking up coins on streets. Santa has a girlfriend named Omitsu (Yasuko Koizumi), but her father Seizo (Kotaro Sekiguchi) does not like having his daughter date a penniless man. He breaks the lovers up and tries to marry his daughter off to a rich man. Desperate, Santa seeks employment and wanders the streets. He can’t find a job at all but has an inspiration. RKO’s King Kong has been released and is a big hit in Tokyo. He decides to capitalize on its success by dressing up as an ape and playing King Kong in a vaudeville theater! He approaches one theater owner to tell him of the idea and the owner is pleased with Santa’s plan, thus giving him the job. Santa’s King Kong show becomes an instant success, with Santa interacting amongst props on the theater stage in his gorilla suit (small buildings, toy airplanes, a doll etc.). One day while performing on stage, Santa sees that Omitsu and her new rich boyfriend are in the audience. Blinded with rage, Santa jumps down from the stage and runs after them - with his gorilla suit on! Santa creates chaos in the town, as firemen and hunters chase him thinking him an escaped gorilla that is running wild in the streets. Eventually Santa confronts the rich boyfriend and knocks him unconscious. He puts the gorilla suit on him and leaves him lying out cold in the street. Just then Koichi comes and tells Santa that the theater owner will give him a lot of money for his performances. Now that he has wealth, Santa gets married to Omitsu.
