A year after the events of Extinction, the Alice clones (Milla Jovovich) attempt to kill Albert Wesker (Shawn Roberts) at the current Umbrella Headquarters, Tokyo. The sole clue is a note left by Solando, which reads: "The law of 4; who is 67?" Shortly after arrival, a storm prevents their return to the mainland for several days. Daniels finds the staff confrontational: the lead psychiatrist, Dr. John Cawley, refuses to hand over records of the hospital staff; Solando's doctor, Dr. Lester Sheehan, had left on vacation after her disappearance, and they are barred from searching Ward C and told that the lighthouse on the island has already been searched, so there is no need to search it. When a patient is being interrogated by Daniels, with a subterfuge she sends Aule away for a few seconds and scribbles "RUN" in Daniels' notepad.

Rod tests Jerry's resolve through a very long telephone conversation while Sugar is able to convince the rest of Jerry's clients to stick with SMI instead. Leaving the office, Jerry announces that he will start his own agency and asks if anyone is willing to join him, to which only 26-year-old single mother Dorothy Boyd (Renée Zellweger) agrees. Meanwhile, Frank "Cush" Cushman (Jerry O'Connell), a superstar quarterback prospect who expects to be the number one pick in the NFL Draft, also stays with Jerry after he makes a visit to the Cushman home. However, Sugar is able to convince Cushman and his father at the last minute to sign with SMI over Jerry. Cushman's father implies they decided to sign with Sugar over Jerry when they saw Jerry attending to Tidwell; an African-American player, versus his son (a white player).

A low-level marijuana dealer named David Clark (Jason Sudeikis) who lives in Denver is robbed of his money and stash, some of which he owes to his wealthy drug lord supplier Brad Gurdlinger (Ed Helms). After David is brought to him, Gurdlinger forces David to smuggle marijuana from Mexico as a solution to clear his debt. Realizing that one man attempting to get through customs is too suspicious, he hires a middle-aged stripper neighbor stage-named Rose (Jennifer Aniston), a runaway and petty thief named Casey (Emma Roberts), and his 18-year-old neighbor Kenny (Will Poulter), to pose as a bogus family called the "Millers". When the Millers reach the compound, they find out that the amount of marijuana they were picking up is not "a smidge" as Gurdlinger had informed them, but actually two tons. Because of the extra load of the marijuana on the RV, one of the radiator hoses breaks while going up a steep incline. A family they had encountered at the border, the Fitzgeralds, consisting of Don (Nick Offerman), Edith (Kathryn Hahn), and Melissa (Molly Quinn), catch up to them and tow the Millers' RV to a repair shop. On the trip to the shop, David learns that Don Fitzgerald is a DEA agent after finding his badge and gun in the glove compartment.
