Ce film déroule une histoire du chemin de fer, depuis les trains à vapeur jusqu'aux années 1930. Sur les routes d'acier loue les services rendus par les cheminots à l'économie française (transport du charbon, de la production agricole...) tandis que défilent des plans d'infrastructures - ponts, tunnels - exaltant la science des ingénieurs ferroviaires. Le corps des cheminots tout entier est salué pour son abnégation et sa rigueur. Le film présente, schéma et graphiques à l'appui, l'organigramme de la SNCF nouvellement créée, avant de se lancer dans la description des œuvres sociales du syndicat des cheminots.

Casey Jones, un conducteur de train à vapeur, doit s'assurer de la bonne livraison du courrier par le rail... Plusieurs problèmes surviennent sur le trajet, tels qu'une inondation consécutive à une tempête (on voit le train cheminer sous l'eau comme un sous-marin), un pont dynamité par un fou (le train s'arrache cependant du fond du canyon en grimpant à la verticale), l'archétypique demoiselle-en-détresse ligotée sur la voie par un assassin, des bandits (que Casey assomme et balance par dessus bord en usant de sa pelle à charbon comme un chasse-mouches) et une vache sur la voie. À toute vitesse, mettant les roues et les rails en fusion, rivetant les fuites de la chaudière debout sur les bielles en plein mouvement, Casey parvient à rejoindre sa destination avec une locomotive en grande partie détruite mais tenant le sac postal.

Donald Kilgore is determined to take a shipment of silk from Seattle to New York City by rail to break a monopoly set up by gangster Wallace Myton. Also aboard the train are Professor Axel Nyberg and his daughter Paula. He is paralyzed (except for the use of his eyes) and needs an operation in New York urgently to save his life. Myton has agents planted on the train to make sure the silk does not arrive in time.

The plot involves an overnight passenger train, the Alpha Trax, which leaves London at midnight and under a full moon. An hour after departure, the train is travelling through a remote and forested part of the English countryside, in a mobile communication "dead zone". Here the train is forced to undergo an emergency brake to prevent a derailment when a deer runs onto the track. Old-timer passenger Ged (Duncan Preston) recalls a similar event happening in the exact same spot - the Great Thornton Forest Rail Crash of 1963 - whereby all the passengers were found dismembered and partially eaten the following morning.
