At the start of the film, the US Government has ordered a branch of the US Military to discontinue tests concerning "the C.H.U.D. project", which is built around the idea that enzymes taken from the sewer dwelling creatures from C.H.U.D. can make hyper-effective killing machines in the army. Jack McCall (Eddie Murphy) is a literary agent who uses his 'gift of gab' to get various book deals, and he's not afraid to stretch the truth to get them. While he is trying to get a book deal from a New Age self-help guru named Dr. Sinja (Cliff Curtis), the guru sees through his deceit and agrees to the deal, only to later deliver a five-page book. That night, a Bodhi Tree magically appears in his backyard. Dr. Sinja goes to Jack's house and they both discover that for every word that Jack says, a leaf will fall off of the tree. When the tree runs out of leaves, the tree will die, as will Jack. In time, he finds that even written words count towards his limit; plus anything that happens to the tree will also affect Jack. When Jack tries to cut it down with an axe, an axe wound appears on him. When squirrels climb the tree, it tickles him. When a gardener tries to poison it with DDT, Jack gets high on the fumes and when the gardener tries to water the tree, Jack starts to sweat/perspire profusely.

Fred Figglehorn, an unpopular, hyperactive and puerile 15-year-old who wears childish dungarees and striped t-shirts, believes himself to be cool and a good singer. Norbit Albert Rice (Eddie Murphy) and Kate Thomas (Thandie Newton) are best friends as children at a combination Chinese restaurant and orphanage owned by their father figure, Mr. Wong (Eddie Murphy), but are separated when Kate gets adopted. While being bullied by twins on a playground one day, Norbit is saved by a tough, overweight girl named Rasputia Latimore (Eddie Murphy) who scares him into being her boyfriend. Though at first she genuinely likes Norbit and cares about him, going as far as intimidating other kids into letting Norbit play with them – Rasputia's kindness deteriorates as she grows up to be an ignorant, arrogant, vindictive, and mean-spirited woman. She eventually pressures Norbit into marrying her, and bullies, insults, and controls him to make sure that she keeps him all to herself. Norbit is also belittled by Rasputia's older brothers, Big Black Jack (Terry Crews), Blue (Lester 'Rasta' Speight), and Earl (Clifton Powell), working as a bookkeeper at their local construction company. The Latimore brothers also run a "security business" and instill fear in everyone in the town except Mr. Wong, who refuses to sell his establishment to them.

Jonathan "Mox" Moxon (James Van Der Beek) is an intelligent and academically gifted backup quarterback for the West Canaan High School football team. Despite his relative popularity at school, easy friendships with other players, and smart and sassy girlfriend Jules Harbor (Amy Smart), Mox is dissatisfied with his life. He wants to leave Texas to go to school at Brown University. He is constantly at odds with his football-obsessed father (Thomas F. Duffy) and dreads playing it under legendary coach Bud Kilmer (Jon Voight), a verbally abusive, controlling authority who believes in winning "at all costs". He has a strong track record as coach, remarking in a speech that "in my thirty years of coaching at West Canaan, I have brought two state titles, and 22 district championships!" On the first day of summer, dim-witted but kind-hearted Ed has a nightmare about talking hamburgers. Troubled by it, he shows up late to work at Good Burger. On the same morning, Dexter, a high school student, steals his mother's car after she leaves for a business trip. He accidentally crashes into his teacher Mr. Wheat's car. With no driver's license or insurance, Dexter is liable to go to jail. Wheat agrees that he will not call the police if Dexter pays $1,900 to cover the damage. In need of money, Dexter gets a job at Mondo Burger, but after a clash with the strict manager, Kurt Bozwell, he is fired. Dexter then finds work at Good Burger, where he meets and reluctantly befriends Ed and a slew of colorful employees.
