En voulant éviter un chien qui traversait la route, Léonard Chaney a provoqué un accident de voiture. Il s'en sort indemne mais pas sa fille, Nancy qui suite à cela, devient aveugle. Chirurgien de renommée, le Dr. Chaney est persuadé qu il doit réparer sa faute en effectuant une greffe des yeux sur sa fille. Ayant réussi des greffes sur des animaux, il compte opérer sa fille. In 1560, several score of Spanish conquistadors, and a hundred Indian slaves, march down from the newly conquered Inca Empire in the Andes mountains into the jungles to the east, in search of the fabled country of El Dorado. Under the command of Gonzalo Pizarro (Alejandro Repullés), the men, clad in half armor, pull cannons down narrow mountainous paths and through dense, muddy jungle. On New Year's Eve, reaching the end of his supplies and unable to go on without more information, Pizarro orders a group of forty men to scout ahead by raft down river. If they do not return to the main party within one week with news of what lies beyond, they will be considered lost. Pizarro chooses Don Pedro de Ursúa (Ruy Guerra) as the commander of the expedition, Don Lope de Aguirre (Klaus Kinski) as his second-in-command, fat nobleman Don Fernando de Guzmán (Peter Berling) representing The Royal House of Spain and Brother Gaspar de Carvajal (Del Negro) to bring the word of God. Also accompanying the expedition, against Pizarro's better judgment, are Ursúa's mistress, Doña Inés (Helena Rojo) and Aguirre's teenage daughter, Florés (Cecilia Rivera, in her only film role).

Au cours de la bataille des Ardennes en 1944, un jeune garçon belge découvre un soldat américain blessé. The film begins with a pre-credits sequence of Secret Agent OSS 117, Colonel Hubert Bonisseur de La Bath (Frederick Stafford) chased by two cars containing enemy agents. OSS 117 manages to blow up both of the vehicles and escape by helicopter. He discovers that his unsuccessful assignment was to obtain information from an agent who had been killed providing information about a mysterious criminal organisation called "The Organisation". OSS 117's superior, Mr Smith briefs him that The Organisation has informed the U.S. State Department that they have a missile that can strike its target undetected and demand $100 Million Dollars as extortion to not use the weapon. As undeniable proof on their intentions and capabilities, they have destroyed an American military base somewhere in Asia presumably with a nuclear weapon.

In Monte Carlo, Marc, a handsome cardsharp, escapes American gangsters who have been ordered to kill him by the boss of a New York gang because he had an affair with the boss's wife. Marc hides in a mission for the poor where Barbara, a wealthy widow, finds him and hires him as her chauffeur. At Barbara's chateau, Melinda, Barbara's niece, becomes attracted to him. Marc discovers that Barbara is hiding her lover, Vincent, in the secret rooms and passageways of the chateau. She and Vincent (a bank robber sought by the police for murdering Barbara's husband) plan to murder Marc so that Vincent may use his passport in escaping to South America. Marc and Barbara begin an affair but are discovered by Vincent, who then kills Barbara but is himself killed by the American gangsters, who mistake him for Marc. Marc and Melinda plan to dispose of the two bodies, but when Melinda learns that Marc is planning to leave without her, she tricks the police into believing Marc guilty and forces him to hide in the chateau's secret rooms. He is her prisoner, just as Vincent had been her aunt's.
