Siddharth (Farhan Akhtar) and Trisha (Vidya Balan) are a happily married couple until Trisha gets pregnant and things start getting complicated. Sid is not ready for a baby as he is just a struggling music composer, yet lies to Trisha. On his friends' advice, Sid starts reading pregnancy books and following Trisha's daily routine by putting a balloon in his shirt. Finally, Trisha gives birth to a baby girl and starts devoting all her time to the baby. Trisha's mother (Rati Agnihotri) feels that Sid is a careless person and can not take this responsibility, at which Sid makes a desperate attempt to behave like a family man but fails, as on one occasion he takes his baby for a walk but forgets her in the market. Frustrated, Sid ask for help from Trisha's brother-in-law Ranveer (Ram Kapoor), who is a typical family man. Ranveer reveals to him that when he gets frustrated, he makes a business trip excuse and stays in a hotel room to enjoy his life as a bachelor. Sid takes on his idea and starts staying in a hotel, with the excuse of working overnight in studio. But soon he has to cut down his idea, due to an added expense at home as Trisha decides to hire an expensive governess (Ila Arun). Sid, then, without telling Trisha, goes for a cheaper solution by shifting to a shared PG along with Manav (Vir Das), a dope-head bachelor. Sid takes Manav's advice and starts living a carefree life.

The film is set in the year 1976, five years after the original film East is East. Little is known about most of the Khan children, except that they seldom communicate with their parents. Tariq is now a hippie (looking like George Harrison) who runs a new age shop with older brother Nazir and has an English girlfriend who is unaware of his true ethnicity, and Maneer is living with his extended family in Pakistan where he is searching for a suitable wife. Sajid, the youngest who no longer wears a parka, is a truant who is constantly bullied due to his Pakistani background, although the headmaster - a former British soldier who had served in the Punjab - is sympathetic and encourages him to embrace his heritage. After Sajid is caught shoplifting, his father George, who has somewhat retained his bullying nature, attacks him at home. When Sajid retaliates by calling him a "dirty Paki bastard," a devastated George states all of his other children in England have become British and he cannot lose Sajid as well, so he decides to take him to Pakistan to meet his extended family and show him that life there is better, though Ella openly disapproves.

Maanav (Arunoday Singh) is a struggling filmmaker who will not compromise on the script he has written. His girlfriend Ruchi (Shahana Goswami), a successful film editor, arranges for him to meet film producer Nitin (Sushant Singh). Nitin likes the script but is not very sure of its box office potential. Maanav then suggests a story from the Panchtantra: A woman is caught red-handed with her lover by her husband and yet, she manages to wriggle out of it Scot-free! Nitin loves the story but finds it too short for a feature film. Maanav then creates three more stories based on the same premise: in a way, the Panchantra story travels in different versions to the modern times through the film. The four stories are woven together by a common story. Mirch itself echoes this structure, with four stories mingling with the main narrative.

The story begins with Col. Krishnakant and his ten men who were sacked from the Indian army as they failed in the China Gate mission. After witnessing the brutal slaying of her Forest Officer father, Sunder Rajan, at the hands of dreaded dacoit Jageera, Sandhya goes to Col. and asks for his assistance in bringing an end to Jageera's rule in the Devdurg region. Krishnakant agrees to assist her and summons ten of his fellow officers and subordinates to assist him in this mission. They assemble at Devdurg with the necessary ammunitions and arms to combat Jageera, not realizing that Jageera has influence over the local police detachment, who will prevent Krishnakant and his men to possess any guns and weapons. This does not deter the men, and they continue to stay on in Devdurg and gain the confidence of the villagers. What the villagers do not know that Krishnakant and his men had all been dishonorably discharged from the army for cowardice and retreating when attacked by the enemy.

Yash Thakur (Sanjay Kapoor) and Suraj Prakash (Salman Khan) are inseparable friends. After completing their studies, they go their different ways. Yash takes over from his father's (Paresh Rawal) underground and mafia-like business, while Suraj becomes an officer of the Central Bureau of Investigation (CBI). Kashi (Sunny Deol) is a kind-hearted wrestler and the dutiful son of Shambhu Nath (Amrish Puri) living in Banaras. Shambhu Nath who was a freedom fighter awarded with Tambra Patra, is an honourable and respectable man in town. Kashi comes to Mumbai with his father for his medical treatment of throat and stays with his brother Shiv Nath (K. K. Raina). Kashi meets Gauri (Meenakshi Seshadri) and starts liking her. He learns that the colony is being terrorized by a tyrannical gangster named Katya (Danny Denzongpa) along with his other six brothers. Before Kashi's arrival, a resident named Sachdev (Om Puri) tries to initiate revolt against Katya but gets ruthlessly killed by him, thereby making Malti (Ila Arun), wife of Sachdev, go mad. Katya and his brothers become enemy with Kashi when he beats up their goons when they were beating up Malti. Shambhu Nath is later diagnosed with throat cancer and has only a few days to live.
