The story centers on a 7-year-old boy, Pepper Flynt Busbee (Jakob Salvati), who is devastated when his father enlists in the army during World War II. The title is a reference to Little Boy, the code name for the atomic bomb dropped on Hiroshima as well as a reference to Pepper's height. Pepper has an extremely close relationship with his loving father. It begins from the very moment he is born when his father cradles him in his arms and mentions how small he is. When Pepper is examined by a doctor the physician is asked if the child is a midget. However, the doctor says that the term is offensive and suggests that he is just a "little boy." However, when World War II begins, Pepper's older brother is declared ineligible for military service because of flat feet and his father joins in his place. Not long after his father leaves for war, Pepper hears the Bible verse, "Truly I tell you, if you have faith as small as a mustard seed, you can say to this mountain, ‘Move from here to there,’ and it will move" (Mt. 17:20), he becomes determined to get enough faith to bring his father home. Nevertheless, as his friend and mentor Father Oliver tells him, faith is useless to anyone who harbors hatred. The film highlights the tremendous hatred against Japanese-Americans that was rampant in America at this time. Father Oliver tells Pepper to befriend a Japanese man named Hashimoto. At first, the racism Pepper has internalized from society makes him hesitant to follow this advice, but as he gets to know Hashimoto, he begins to value their friendship and learns from Hashimoto to stand up to those who bully him about his height. The plot then takes a turn when Pepper's father is reported missing in action. Pepper's mother (Emily Watson) struggles to keep her family together in the face of these overwhelming hardships.

Budget cutbacks at the school jeopardize the continuation of its music program, which would result in its teacher, Marty (Henry Winkler), being laid off. Concerned for both his colleague and his students, Scott attempts to raise the $48,000 necessary to keep the music program alive. At first, he works as a night instructor for a citizenship class. One of his students, Niko (Bas Rutten), approaches him to get some outside tutoring and Scott reluctantly agrees. When Scott arrives at Niko's apartment, he realizes that Niko was a former mixed martial arts (MMA) fighter. While watching the UFC at Niko's apartment, Scott learns that the loser of the fight receives $10,000, which gives him the idea of raising the money by fighting and losing in MMA himself. Niko begins training him in defense, and later brings him to another trainer, Mark (Mark DellaGrotte), to train for offense, after a knockout on Scott's behalf he realized he could win more and fight less. While Mark trains with Scott, Malia De La Cruz (Charice Pempengco), one of Scott's students and a band member, helps Niko study for his citizenship test by putting the information into songs. In 1978, five childhood friends win their junior high school basketball championship. During their celebration at a rented lake house, the friends' coach, Bissau (Blake Clark), encourages them to live their lives in a similar way to how they played the game. In 2010, Lenny Feder (Adam Sandler) is an ambitious Hollywood talent agent who is married to fashion designer Roxanne Chase (Salma Hayek), and has three children—one daughter Becky and two sons, Greg and Keith (Jake Goldberg and Cameron Boyce); his sons have become very spoiled much to his annoyance. Eric Lamonsoff (Kevin James) claims he is now a co-owner of a lawn furniture company, is married to Sally (Maria Bello) and has two children, Donna and Bean (Ada-Nicole Sanger and Morgan Gingerich). Much to Eric's chagrin, Sally continues to breastfeed Bean. Kurt McKenzie (Chris Rock) is a stay-at-home father who is married to Deanne (Maya Rudolph), the primary breadwinner of the family, and has two children, Andre and Charlotte (Nadji Jeter and China Anne McClain). Deanne is pregnant with another child and her mother (Ebony Jo-Ann) also lives with the family. Rob Hilliard (Rob Schneider) has been divorced three times and has daughters Jasmine, Amber, and Bridget (Madison Riley, Jamie Chung, and Ashley Loren) from those marriages. His current wife, Gloria (Joyce Van Patten), is 30 years older than him. Marcus Higgins (David Spade) is a slacker and lothario. All five friends regularly harass each other in comedic fashion throughout the film: Lenny for being rich; Eric for being overweight; Kurt for being skinny and not being more useful; Rob for his way of saying "Maize!" and for having a much older wife; and Marcus for being sexually juvenile.
