Six ans après les événements du premier film (Les Mondes de Ralph), Ralph La Casse et Vanellope restent toujours les meilleurs amis du monde et s'amusent chaque nuit à jouer ensemble. Alors que Ralph est heureux de la vie qu'il a, Vanellope avoue être ennuyée par son jeu, qui est trop prévisible, et aimerait de la nouveauté. Un jour, Ralph essaie d'exaucer son souhait en créant une course bonus dans l'un des circuits. Vanellope va contre la joueuse de la borne d'arcade pour profiter de la course à fond, tant et si bien que la joueuse, en forçant trop sur le volant, le casse. La situation paraît désespérée lorsque le propriétaire de la salle d'arcade, M. Litwak, annonce que le créateur de Sugar Rush a fait faillite, quand l'un des adolescents trouve un volant sur eBay. Deux élèves en CM1 à l'école Jerome Horwitz (ou Albert Martin en français) de Piqua (Ohio), Georges Glousse (le scénariste) et Harold Golade (le dessinateur) créent des bandes dessinées dans leur cabane et multiplient les blagues à l'école. Mais un jour, après les avoir pris sur le fait grâce à une caméra de surveillance cachée dans une tortue-jouet, M. Chonchon, leur proviseur hargneux, décide de les séparer en les plaçant dans deux classes différentes. Pour échapper à cette séparation, ils hypnotisent, grâce à une hypno-bague trouvée dans un paquet de céréales, M. Chonchon, et lui font croire successivement qu'il est un poulet, un singe puis le Capitaine Superslip, un superhéros de leur création. Quand il entend un claquement de doigts, M. Chonchon enlève ses vêtements et sa moumoute. Vêtu d'un slip immaculé, il se fait une cape d'un rideau rouge, lève le poing gauche, l'index pointé en l'air et lance le fameux slogan du Capitaine Superslip : « Tra-La Laaaa ! ». Blue macaws Blu and Jewel, and their three children, are living happily in the city until Jewel realises that her children are becoming more like humans. Elsa, Princess of Arendelle, possesses cryokinetic powers, with which she is able to produce or manipulate ice, frost and snow at will. A mysterious vehicle using a huge magnet steals a highly potent mutagen known as PX-41 from a secret laboratory in the Arctic Circle. The Anti-Villain League (AVL) tries to recruit former supervillain, Gru, to find who stole the mutagen, but Gru refuses, claiming he is a legitimate businessman now as well as the father of Margo, Edith, and Agnes. Dr. Nefario, Gru's friend and assistant, has decided to leave him for new employment, claiming he "missed being evil." Gru reluctantly partners with undercover AVL agent Lucy Wilde, and they search a local mall, the Paradise Shopping Mall, disguised as bakery employees. Mexican restaurant owner, Eduardo Pérez, is suspected by Gru of being "El Macho", a legendary supervillain who supposedly died after skysurfing a shark into the center of an active volcano. Gru and Lucy break into Eduardo's restaurant at night, but find nothing to prove that he is the culprit. Meanwhile, Agnes expresses her wish to have a mother someday and believes that Gru will fall in love with Lucy, but despite her excitement, Gru tells her that his relationship with Lucy is strictly professional.

Ted Wiggins is an idealistic 12-year-old boy [citation needed], who lives in "Thneedville", a walled city that, aside from the human citizens, is completely artificial; everything is made of plastic, metal, or synthetics. Ted has a crush on local environmentalist Audrey, who wants to see a "real tree" more than anything in the world, and decides to find one in order to impress her. His energetic Grammy Norma secretly tells Ted the legend of the Once-ler, who will tell anyone about trees if you brought him fifteen cents, a nail, and a shell of a great-great-great grandfather snail. When Ted leaves Thneedville in search of the Once-ler, he discovers that the outside world is a contaminated, empty wasteland. Once the boy finds him, the Once-ler agrees to tell Ted about the trees on the condition that he listens to the story over multiple visits. Ted agrees, but on his way home, he encounters the mayor of Thneedville, Aloysius O'Hare, who is also the proprietor of a company that sells bottled oxygen to Thneedville residents. O'Hare explains to Ted that because trees produce oxygen free of charge, he considers it a threat to his business whenever he hears people talking about them. After revealing that he has "security camera eyes" all over the city, O'Hare pressures Ted to stay in town. However, Ted continues to sneak out of O'Hare's sight (with his grandmother's encouragement) and learns more of the trees' history.

When Litwak's Family Fun Center & Arcade closes at night, the various video-game characters leave their normal in-game roles and are free to travel to other games. Within the game Fix-It Felix, Jr., the characters celebrate its titular hero but loathe the game's villain character, Wreck-It Ralph. At a support group for video-game antagonists, Ralph reveals his desire to stop being the bad guy. Back home, Ralph finds the other characters celebrating their game's 30th anniversary without inviting him. In 1837, the Pirate Captain (Hugh Grant), inexpert in the ways of pirates, leads a close-knit, rag-tag group of amateur pirates who are trying to make a name for themselves on the high seas. To prove himself and his crew, the Pirate Captain enters the Pirate of the Year competition, with the winner being whoever can plunder the most. After several failed attempts to plunder mundane ships, the Pirate Captain talks with his best friend and first mate, the Pirate with a Scarf (Martin Freeman), about giving up and selling baby clothes as a living, but is convinced to try boarding the next ship they find, which happens to be the Beagle. They find no treasure but succeed in capturing Charles Darwin (David Tennant).
