Keith Lemon (Leigh Francis) is an aspiring business entrepreneur from Leeds, who is trying to secure investment for his invention, the Securipole (a metal pole put in the driveway to prevent car theft). In December 1941, just before the USA's entry into World War II, an American agent from the Naval Intelligence Office (Cusack) arrives in Shanghai to find his friend Conner (Jeffrey Dean Morgan) was recently murdered. Determined to find out who killed Conner and why, he begins working at the Herald using the alias Paul Soames, a Nazi-sympathizer cover he used while stationed in Berlin. He meets Anthony Lan-Ting (Chow Yun-Fat) an influential crime lord and Captain Tanaka (Ken Watanabe) at the German embassy in Shanghai, during an invitation only event. He later befriends Anthony when he saves him during an attack on Japanese officers by the Chinese Resistance at a night club.

Mike Enslin (John Cusack) is a cynical, skeptical author who, after the death of his daughter Katie (Jasmine Jessica Anthony), writes books appraising supernatural events in which he has no belief. After his latest book, he receives an anonymous postcard depicting The Dolphin, a hotel on Lexington Avenue in New York City bearing the message, "Don't enter 1408." Viewing this as a challenge, Mike forces the hotel to allow him to book the room, referencing a law that any hotel room in New York can be requested as long as it meets safety standards. The hotel manager, Gerald Olin (Samuel L. Jackson) tries to dissuade Mike from checking into the room, explaining that 56 people have died in the room over the past 95 years, and that no one has lasted more than an hour inside it. Mike, who does not believe in the paranormal, insists on staying in the room, and asks Olin if he thinks it is haunted; Olin replies that it is "evil" rather than haunted.

With the exception of the opening and final scenes, which depict the 1941 suicide by drowning of Virginia Woolf in the River Ouse, the action takes place within the span of a single day in three different years and alternates between them throughout the film. In 1923, Virginia has begun writing the book Mrs Dalloway in her home in the town of Richmond outside London. In 1951, troubled Los Angeles housewife Laura Brown escapes from her conventional life by reading Mrs Dalloway. In 2001, New Yorker Clarissa Vaughan is the embodiment of the novel's title character, as she spends the day preparing for a party she is hosting in honor of her former lover and friend Richard, a poet and author living with AIDS who is to receive a major literary award. Richard tells Clarissa he has stayed alive for her sake, and the award is meaningless because he didn't get it sooner, until he was on the brink of death. She tells him she believes he would have won the award regardless of his illness. Richard often refers to Clarissa as "Mrs. Dalloway" - her namesake - because she distracts herself from her own life the way the Woolf character does.
