When visiting San Francisco, Tony Patucci, an ageing illiterate winegrower from the Napa Valley, sees waitress Amy Peters and falls in love. Returning home, he persuades his foreman Joe, an incorrigible womanizer, to write her a letter in Tony's name. Tony's courtship by mail culminates with a proposal, and when she requests a picture of him, he sends one of Joe. Amy accepts and goes to Napa to be married. Although horrified to discover that her prospective husband is the portly Tony, she decides to go through with the marriage. However, while Tony is in bed after an accident, Amy and Joe have an affair. Two months later, as Tony plans the wedding, she discovers that she is pregnant. Upon learning this, Tony pummels Joe, who leaves the vineyards. but forgives Amy, and insists that they still be married, But she is unable to forgive herself, so she leaves with the priest who's come to marry them, while Tony looks on, hoping that she will return one day.

The musical's story spans about forty years, from the late 1880s to the late 1920s. Magnolia Hawks is an eighteen-year-old on her family's show boat, the Cotton Palace which travels the Mississippi River putting on shows. She meets Gaylord Ravenal, a charming gambler, falls in love with him, and eventually marries him. Together with their baby daughter, the couple leaves the boat and moves to Chicago, where they live off Gaylord's gambling winnings. After about ten years, he experiences an especially bad losing streak and leaves Magnolia, out of a sense of guilt that he is ruining her life because of his losses. Magnolia is forced to bring up her young daughter alone. In a parallel plot, Julie LaVerne (the show boat's leading actress, who is part African-American, but "passing" as white) is forced to leave the boat because of her background, taking Steve Baker (her white husband, to whom, under the state's law, she is illegally married) with her. Julie is eventually also abandoned by her husband, and she becomes an alcoholic. Magnolia's becomes a success on the stage in Chicago. Twenty-three years later Magnolia and Ravenal are reunited at the theater in which Kim, their daughter, is appearing in her first Broadway starring role.

As described in a film magazine, Betsy O'Hara (Barrymore) marries Lord Frederick Berolles (Ratcliffe) to please her parents, and while now is Lady Frederick, she finds herself miserable when her husband constantly insults her after discovering that she loves another. It does not help that Sir Paradise Fuldes (Herbert), the man she would have married except for his poverty, soon thereafter comes into a fortune. Her sister Kitty (Childers), while married to a man of position, has foolishly compromised herself with Robert Montgomery (Kilgour), a cad who forces her to visit his rooms on threat of disclosure. While assisting her sister in the recovery of some old love letters, she is surprised in the apartments of Montgomery, and assumes the situation to save her sister's reputation. Lady Frederick does not encourage him, but he will not take no for an answer. His wife Lady Mereston (Gordon) appeals to her brother Sir Paradise to intervene, and while Lady Frederick resents the interference, she sends Mereston about his business using an ingenious device, and finds her reward in the arms of Paradise.

As described in a film magazine, Esther (Barrymore), upon the death of her father, is advised by her kindly neighbors to get married. She is forced to sell the homestead and marries a domineering old miser named John Martin (Carrington), who lives with his maiden sister Ruth (Allen). Because she passes the time of day on the street with young men, her husband becomes jealous. He chokes her after he finds a letter from a former friend, Dr. Henry Grey (Hale), and she decides to leave him. While escaping with her son she is detained in a hut by a drunken farmer who tries to embrace her. She shoots him dead, and a posse arrests her for the death of her husband, who was found strangled in the library. However, the death confession of the real murderer clears her of her husband's death, and the return of her former friend from France completes her happiness.

As described in a film magazine, Clorinda Gildersleeve (Barrymore) has drifted into an affair with a married man, Leslie Palliser (Ellis). She goes to Europe to forget and meets Malcolm Grant (Davidson), a young surgeon, who falls in love with her. However, because of her affair with Palliser, she believes she has no right to accept his love. She hears an excellent sermon preached by Reverend Arthur Bainbridge (Gillmore) on the text "Woman, go and sin no more" and appeals to the minister in regards to Malcolm's attentions. She sends Malcolm to see the minister, but he does not disclose Clorinda's secret. The minister interests her in a home for erring girls and Clorinda adopts one of the inmates. Reverend Bainbridge urges her to marry Malcolm, but during the ceremony she falls in a faint. That night, however, she writes to the minister announcing her marriage to Malcolm and their departure for France, where he will do surgical work and she will become a nurse.
