Andy Brewster (Seth Rogen) is a UCLA-graduate organic chemist and inventor. He is attempting to get his environmentally friendly cleaning product, ScioClean, in a major retail store. However, each retail store he visits dismisses him before he can end his pitch. After a disappointing sales pitch to K-Mart, he visits his mother, Joyce Brewster (Barbra Streisand), in New Jersey before leaving on a cross-country trip to Las Vegas, lying to her that his pitch ended well so she won't worry about him. While there she reveals to him that he was named after a boy she fell in love with in Florida named Andrew Margolis, whom she hoped would object to her marriage with Andy's father. However, he never did and she felt that she never mattered to him afterwards. After a little research, he finds Andrew Margolis is still alive and unmarried living in San Francisco. He invites his unknowing mother on the trip, claiming he wants to spend some time with her.

Jan Paul Beahm (Shane West) grows up in Los Angeles through a troubled childhood; he does not know his biological father, his mother is an alcoholic, and his older brother dies from a heroin overdose. He is an avid reader and develops into a "frighteningly intelligent" student at University High School, where his antisocial behavior leads the administration to give him straight A's if he agrees not to return. In December 1975, at age 17, he proposes to his friend Georg Ruthenberg (Rick Gonzalez) that they start a band, showing him potential lyrics and claiming to have a "five-year plan" inspired by the David Bowie song "Five Years". They recruit Terri Ryan (Bijou Phillips), Belinda Carlisle (Lauren German), and Becky Barton (Amy Halloran) for the group and con money for instruments. Jan Paul comes up with the name Germs, representing the germination of an idea. He sings while Georg plays guitar, Terri plays bass guitar, and Becky plays drums.

Doug Stauber (Seann William Scott) is the assistant manager of a branch of Donaldson's, a supermarket chain in Chicago. Slick salesman Jimmy Starks (Pearce) has auto problems in a small New Mexico town and while his car is in the shop he visits low-rent fortune teller Vacaro (J.K. Simmons) to pass the time. The supposed seer tells him he will have good fortune soon, but looking deeper relates the information that his future is blank, and he is safe only until the first snow of winter beyond which there is no future to foretell. The act upsets Jimmy and rekindles old transgressions and makes him feel he is on a collision course with destiny especially when an old friend Vincent (Shea Whigham) returns from a jail sentence that he went to in Jimmy's place after a bad business deal. Jimmy becomes obsessed with knowing more of his future and re-visits Vacaro but the old man can only tell the salesman that he has related to him all he can see. Jimmy feels that Vincent has returned to kill him and that he must do something to change the course of his future but Vacaro convinces him to accept his fate.
