The film is a satire of the women in prison film genre and deliberately implements many of the tropes commonly found in such films. Such scenes include nude shower scenes, fight scenes, and a suggested romantic relationship between one of the inmates and an administrator. The overall plot involves a new influx of girls coming to the school. They are immediately confronted with Charlie Chambliss (Williams) who is the de facto leader of the school and has an exceedingly close relationship with the head of the ward, Edna (Ast). Charlie and her circle are given special privileges by Edna and it is suggested that Charlie and Edna enjoy a more intimate relationship. Charlie runs a secret society of girls who are loyal to her and to whom she offers protection. The two main new girls break several of Edna's rules and are punished decisively for their infractions.

The film begins with a young teen girl screaming, only to reveal that she is a student at a frat party. While at the party, the camera cuts to Peter, who is the president of Alpha Sigma Rho. He decides that four new pledges should have an initiation. He decides that the group of youths: bookish Marti, brawn Jeff, partying Denise, and surfer Seth, should spend the night in the supposedly "haunted" estate, Garth Manor, where there was a murder twelve years ago prior to the present events. Peter began telling the story about the previous events that took place twelve years earlier as he leads them to the estate. The house's former owner, Raymonde Garth, who dismembered his family in the parlor strangled & killed his wife to death & kills his four deformed children: Margeret, Morris, & Suzanne, in seemingly violent ways, before finally hanging himself. However, the youngest child, Andrew Garth, dead or alive, was never found and is rumored to have seen the murders and fled the scene. Rumor has it that the deformed Andrew might still be living inside Garth Manor and his remains are left hidden as a deformed monster.

Penelope, a young hairdresser, discovers her vagina can talk when it criticizes a lover's performance, who leaves in a huff. At the salon where she works, her talking vagina insults a lesbian client, which leads to her being fired. Penelope goes to a psychiatrist, Dr. Pearl, where she reveals her "problem". In the psychiatrist's office, her vagina reveals a new talent, singing. It has a propensity for singing show tunes. Dr. Pearl reveals her secret to friends of his in show business. At a meeting of the American Medical Association, Penelope and her talking vagina, now called "Virginia", are revealed to the public for the first time. Virginia regales the assembled physician with show tunes. Dr. Pearl becomes her agent, and over Penelope's objections, launches Penelope and Virginia on an entertainment career. At a show emceed by Professor Irwin Corey, Virginia sings in public for the first time, becoming a star after crooning a disco tune. Virginia increasingly becomes the tail that wags the dog, with Penelope becoming increasingly unhappy as "they" become a successful act on a cross-country tour. Despite her new success, Penelope decides to kill herself until she sees the lover from the start of the movie and discovers that he has a talking penis.
