The story focuses on Kishen (Akshay Kumar), a well-to-do businessman, who is unhappily married to Mala (Aishwarya Rai). They have a grown-up son, Bunty (Aditya Roy Kapoor), who does not want to get married because of the loveless relationship between his parents. Bunty's girlfriend, Tanya (Sudeepa Singh), keeps proposing to him but he always rejects. On their 35th anniversary, Kishen and Mala get in an argument because of her friend Kundan (Rannvijay Singh). Kundan had been in love with Mala for 30 years and still dislikes Kishen. This leads to a fight between Kishen and Kundan. Indian-born Manmohan Malhotra (Rishi Kapoor) re-located to London, England, established himself, returned to India, got married to Bebo (Nina Wadia), and after four years got a visa for her so that she could live with him. Shortly thereafter, she gave birth to Jasmeet (Katrina Kaif). Manmohan was always embarrassed by Bebo as she was fat and could not speak English. Obsessive and temperamental bank manager Vijay Singh Rajput (Bachchan) loses his job after badly beating up a bank clerk who tried to embezzle money. It is a romantic-drama/comedy film. Subhashree portrays an N.R.I girl living in London whose father (portrayed by Biswajit Chakraborty) wants her to get married with an Indian lad. However, Subhasree's character is deeply involved in a yet-platonic relationship with a half-Brit boy Harry. Subhasree's brother (played by Tota Roy Choudhury) is involved in a relationship with a British girl and wants to marry her. However, his father refuses to let him marry the girl and this event promptly induces him to leave his parents' home. Meanwhile, Subhasree's father manages to bring his wife and daughter to India where he can find a suitable groom for his daughter. This is where they meet Dev, a quintessential charming Bengali lad who falls head-on in love with Subhasree and their marriage is invariably finalized since Dev's father is an old friend of Subhasree's Dad as well. However, although Subhasree evidently likes Dev, she hatches a plan to delay and finally, annul the engagement by implementing her brother's plan of bringing Dev to London and 'breaking up' with him abroad. Poran Jai Jolia Re is the story of how Dev manages to win his love over all adversities in a foreign land.

The movie opens in an engineering college in Ooty with a narration of the characters. Rajiv Samuel (Abbas) is a computer engineering student and is one of the top students in the college, whereas Rajesh (R. Madhavan) is a mechanical engineering student who is the bad guy of the college. A mutual problem between the two groups led by both Samuel and Rajesh puts both Sam and Rajesh at loggerheads and they pull pranks on each other to get the other in trouble. It culminates into a fist fight but is soon broken up their friends. In the fight, Rajesh punches Sam in the face, which is against the rule, and Sam promises to return the favor in future. They graduate from the college soon and promise to not see each other ever again. Sam goes abroad for job, while Rajesh stays back in India saying that he does not want to leave his motherland.
