Akeelah Anderson, an 11-year-old spelling enthusiast, attends Crenshaw Middle School, a predominantly black school in South Los Angeles. She lives with her widowed mother, Tanya, her older sister Kiana, her older brothers Devon and Terrence, and her infant niece. Her principal, Mr. Welch suggests that she sign up for the Crenshaw Schoolwide Spelling Bee. She follows his advice and ends up winning. Soon after, Dr. Joshua Larabee, a visiting English professor and Mr. Welch's friend from college, tests Akeelah and decides that she is good enough to compete in the National Spelling Bee. Nevertheless, Dr. Larabee declines to coach her because she is rude to him. As a result, Akeelah studies on her own to prepare for the district spelling bee. Although Akeelah misspells her word during the final round of the bee, she qualifies for the regional bee when her sister Kiana catches the other finalist cheating. She also meets and befriends Javier Mendez, a 12-year-old Mexican American boy and fellow speller. Javier invites her to join the spelling club at his Woodland Hills middle school.

Joe Scheffer (Tim Allen) is a recently divorced single parent, and a talented audio/visual specialist at STARKe Pharmaceuticals, his place of employment. One day Joe pulls into the parking lot at work to find his co-worker Mark McKinney (Patrick Warburton) parking in a spot that has been reserved for those who have worked for the company for 10 years. However, McKinney has only worked there for 7 years. When Joe confronts McKinney about this, McKinney assaults him in front of his young daughter Natalie (Hayden Panettiere). Joe falls into a state of depression until Meg Harper (Julie Bowen), the Wellness Coordinator at STARKe, accidentally ignites a fuse in him when, in a fit of frustration, she asks Joe, "What do you want?" Joe is suddenly stirred to action by this question, and decides he wants a rematch to reclaim his dignity and self-respect, which he felt McKinney took from him.

As the U.S. Subic Bay naval base's operations slowly wind down and naval manpower begins to dwindle, Commander Hamilton (Wolfgang Bodison) relies on three U.S. Navy Seals to help keep the base secure. James Dunn (Wayans), a United States Marine who served in the Gulf War, is wrongly accused of an assassination of an officer he had disputed with. Dunn is later saved from death row and recruited for top-secret special operations squad led by Lt. Col. Grant Casey (Voight). Their mission is to neutralize criminals who had avoided conventional law enforcement methods. Vanessa Lutz (Witherspoon) is a poor, illiterate teenage girl living in the slums of Los Angeles. After her mother, Ramona (Amanda Plummer), is arrested in a prostitution sting, she runs away with a car stolen from her social worker guardian to stay with her grandmother in Stockton. Along the way, Vanessa stops to see her boyfriend and classmate Chopper Wood (Bokeem Woodbine), a local gang member, to tell him about her excursion and he gives her a gun to sell upon arriving at her destination. Minutes after Vanessa leaves Chopper, is killed in a drive-by shooting by rival gang members. The film covers the court-martial of two U.S. Marines, Lance Corporal Harold Dawson and Private Louden Downey, who killed a fellow Marine, Private William Santiago, at the Guantanamo Bay Naval Base in Cuba. Santiago compared unfavorably to his fellow Marines, had poor relations with them, and failed to respect the chain of command in attempts at being transferred to another base. An argument evolves between base commander Colonel Nathan Jessup and his officers: while Jessup's executive officer, Lieutenant Colonel Matthew Markinson, advocates that Santiago be transferred immediately, Jessup regards this as akin to surrender and orders Santiago's commanding officer, Lieutenant Kendrick, to train Santiago to become a better Marine.
