Left off after the events of the first movie The Blob, an oil pipeline layer named Chester (Godfrey Cambridge) returns to his suburban Los Angeles home from the North Pole, bringing with him a small sample of a mysterious frozen substance uncovered by a bulldozer on a job site. Prior to taking the blob to a lab to be analyzed, he places the storage container with the substance in his freezer, but he and his wife (Marlene Clark) accidentally let it thaw, releasing "the Blob". It starts by eating a fly, then a kitten, Chester's wife, and then Chester himself (while he is watching a television broadcast of the film The Blob).

In London, a serial killer is raping women and strangling them with neckties. Most of the film takes place in Covent Garden, which at the time was still the wholesale fruit and vegetable market district. Fairly early in the film, the audience sees that fruit merchant Robert Rusk (Barry Foster) is in fact the murderer. However, circumstantial evidence has already built up around his friend Richard Blaney (Jon Finch).

The film focuses on the trials and tribulations of Alexander Portnoy, a Jewish man employed as the assistant commissioner of human opportunity for New York City. Employee John David Welles attempts to steal rocket booster plans from the Groundstar facility. His attempt goes awry and he is badly disfigured in an explosion and barely escapes. He stumbles to the home of Nicole Devon, and collapses. She calls an ambulance, the authorities are alerted, and soon Welles is operated on, given plastic surgery and interrogated by a gung-ho government official named Tuxan. But Welles claims to have no memory of his crime. In fact, he claims no memory of his life at all, save for brief glimpses of a woman and small boy frolicking on a beach.
