La Moustache opens with Marc Thiriez, a middle aged Parisian, taking a bath and asking his wife if he should shave off the moustache he has sported for most of his adult life. His wife, Agnès, wryly comments that she wouldn't recognize him without it, yet as she leaves, Marc shaves the moustache off. Upon her return, Agnès is angry Marc did not let her in the door when she rang. Marc lies and says he broke his shoelace. In the car Marc asks his wife if she notices anything. She does not respond, wanting to know if he thinks she is dressed too sexy for the party. Later that evening, as the two visit their friends Serge (Agnes' first husband) and Nadia, Serge tells a funny story from his marriage to Agnès, how she was sneaky and refused to admit it, even when found out. Agnès again denies the sneakiness. Frustrated that Serge and Nadia also do not notice that Marc has shaven his trademark moustache, Marc fights with Agnès in the car as they return home, frightening Agnès. As they lie in bed that night, another fight over the moustache ensues as Agnès digs in her heels and tells Marc he's never had a moustache and that she fears for his sanity. Agnès phones Nadia and Nadia claims Marc has not had a moustache in fifteen years. Agnès takes a sleeping pill to sleep.

Michelle Rosen, an introverted and insecure high school student, is approached at lunch by popular jock Aaron Cussler, who invites Michelle to a party he is throwing. Charmed by Aaron, Michelle goes to the gathering, where she makes out with Aaron (to the annoyance of Aaron's occasional girlfriend Debbie) and befriends another student named Wendy, a formerly overweight outcast who is now a member of Debbie and Aaron's clique. As the night progresses, Aaron and two of his friends (one them Wendy's boyfriend, Steve) get Michelle drunk, and slip her a roofie. Aaron and Jake then rape Michelle (at one point violating her with a bottle) in a bedroom while Steve, who had grown reluctant, restrains her. Afterward, the boys' girlfriends find the unconscious Michelle, and convinced by Aaron that Michelle was the aggressor, Debbie uses packing tape to tie Michelle to a tree in the backyard, after writing "Slut" and "Ho" on her. The only one who tries to aid Michelle is Wendy, but she is made to back off by Jake's girlfriend Heather, under threat of being made a pariah. Michelle eventually frees herself and goes home, where she engenders no sympathy from her emotionally abusive mother.

The film is split into three main segments spanning thirty years. The first segment, in 1976, begins with the return of an artist, Gengnian to his wife, Xiuqing and son after several years of re-education during the Cultural Revolution. Focusing on the Madagacar penguins and taking place before the events of the first Madagascar, the youngest penguin on the team, Private, slips out of the zoo on Christmas Eve to find a present for a lonely polar bear named Ted. While roaming the streets of Manhattan, he is captured by Nana (the mean old lady from the first film) who mistakes him for a chew toy for her vicious dog, Mr Chew. The other three penguins, Skipper, Kowalski, and Rico, rescue Private from Nana's apartment before it is too late. They escalate into chaos against Mr. Chew, all-the-while, not noticed by Nana, who is occupied watching a football game. When they are done, they detonate the door, which is a running gag in the film, finally attracting Nana's attention and leaving Mr. Chew to take the fall for what the penguins have done to her place.

The film focuses on several characters. One of the main protagonists, Dean (Justin Long), has been a waiter for four years since graduating from high school, and has not earned a degree during his four years at a community college. When Dean learns from his mother that a former high school classmate, Chett (Travis Resor), now has a bachelor's degree in electrical engineering, he begins to rethink his direction-less life. Dean's lecherous friend and co-worker, Monty (Ryan Reynolds), is in exactly the same situation, but has accepted that his future lies with the restaurant, Shenaniganz. Monty is put in charge of training Mitch (John Francis Daley), a newly hired waiter who is constantly interrupted throughout most of the film before he can speak, usually by Monty. Also working with Dean is Calvin (Robert Patrick Benedict), a hopeless romantic who cannot urinate in public, and Dan (David Koechner), the uptight manager who is grooming Dean as his protégé. Rounding out the staff are 17-year-old hostess Natasha (Vanessa Lengies), abrasive waitress Naomi (Alanna Ubach), waitress (and Monty's ex-girlfriend) Serena (Anna Faris), Dean's girlfriend and fellow waitress Amy (Kaitlin Doubleday), stoner busboys T-Dog and Nick (Max Kasch and Andy Milonakis), head chef Raddamus (Luis Guzman), the insane, unsanitary chef Floyd (Dane Cook), lesbian bartender Tyla (Emmanuelle Chriqui), and the pensive and philosophical dishwasher Bishop (Chi McBride).
