Dale Denton (Seth Rogen) is a 25-year-old process server and habitual pot smoker. He makes a visit to the home of his drug dealer, Saul Silver (James Franco), to buy weed. Saul tells him that he may already know the identity of Dale's next service, Ted Jones (Gary Cole). Dale drives to Ted's house and witnesses Ted and a cop, Officer Carol Brazier (Rosie Perez), shoot an Asian man to death. Dale panics and flees the area, but leaves his roach at the scene which contains a rare strain of marijuana called Pineapple Express. Ted is able to identify the strain and sends his two henchmen, Budlofsky and Matheson (Kevin Corrigan and Craig Robinson) to dealer Red (Danny McBride), who tells them that he has only sold the pot to Saul.

Kai and Stefan are two friends who own a gourmet pizza delivery business as a front for selling cannabis. In 1971, a policeman catches Elmo McElroy, a recent college graduate with a degree in pharmacology, smoking marijuana. Because of his arrest and conviction, he is unable to find work as a pharmacologist. In the present day, a drug lord called "the Lizard" calls a meeting of his organisation, hoping to sell a brand new substance invented by Elmo. The meeting goes badly when Elmo, in a bid to escape from the Lizard's control, blows up the building, killing everyone but the Lizard. After his best friend Mike dies of a drug overdose, San Fernando Valley businessman Jack Magnus spirals into depression and drug addiction, losing his job, getting a severe infection on his left arm, and becoming homeless, forcing him to live in the warehouse where he works as a security guard. One night, a pair of dealers Jack owes money to break into the warehouse, and decide that since Jack does not have any money, to rob the building, and kill Jack by injecting him with a mixture of PCP, heroin, and crystal meth. Instead of killing Jack, the cocktail of drugs causes him to have a psychotic break, and drilled by paranoid delusions and a hallucination of Mike, he kills the dealers with an electrical jackhammer that was among the tools in the warehouse machine shop.
