On Christmas Eve, 1989, aspiring filmmaker Mark Cohen, and his roommate, Roger Davis, learn that the rent previously waived by their former friend and landlord, Benjamin “Benny” Coffin III, is due. A straight bass player in an "all gay band" called The Jerk-Offs, Nick O'Leary (Michael Cera) is a teenager from Hoboken, New Jersey. Nick is still heartbroken after his girlfriend, Tris (Alexis Dziena), broke up with him three weeks, two days, and 23 hours ago, and continues to make for her "breakup" mix CDs. Thom (Aaron Yoo) and Dev (Rafi Gavron), his gay bandmates who both hate Tris, convince Nick to perform at a club because his favorite band— the legendary, publicity-shy indie band Where's Fluffy? —is performing somewhere in New York City that night. Norah Silverberg (Kat Dennings) attends the same high school as Tris, Convent of the Sacred Heart, and though Norah and Tris dislike each other, they have a mutual friend named Caroline (Ari Graynor). Norah has not met Nick, but shares his taste in music based on recovering his mix CDs after Tris discards them in the trash. That night, the three girls end up at Arlene's Grocery on Manhattan's Lower East Side, where The Jerk-Offs are playing. Norah asks Nick to pretend to be her boyfriend to show up Tris. Norah kisses him, unaware that he is the ex-boyfriend of Tris. Caroline gets drunk and Norah wants to take her home, but Nick's bandmates (who see Norah as a potential new girlfriend for him) offer to take Caroline home so Norah and Nick can spend time together trying to find the show.

The film's plot revolves around Kanthaswamy (Vikram), who is an Income Tax officer in the CBI, and his friends, granting the wishes of the needy who write letters to a local Lord Muruga temple by disguising himself as an anthropomorphic rooster. During a CBI income tax raid, he discovers a stash of black money owned by Ponnusamy (Ashish Vidyarthi), a rich and corrupted entrepreneur. Ponnusamy pretends to be paralysed to escape the questioning. Angered by the news that her father is paralysed, Ponnusamy's daughter, Subbulakshmi (Shriya Saran) goes after Kanthaswamy to seek revenge by pretending to be in love with him. Kanthaswamy, knowing Subbulakshmi's real intentions plays along in the game. This forms a cat and mouse chase between the two. In a comical role, Thengakadai Thenappan (Vadivelu) is also sought after by a local police inspector who tries to solve the mystery of "Lord Muruga saving the people". The fate of Kanthaswamy's disguise leads into the climax of the story. The local police inspector was able to bring out the truth that a CBI is involved in the role behind the secret work of helping the needy people although he was not able to confirm it. Kanthaswamy finds the password of Subbulakshmi's family bank account, in which the black money is illegally saved. He tricks Subbulakshmi into revealing the password and answers for the security questions indirectly. Kanthaswamy finally transfers the money to his account which would be used for helping the poor and needy people. Ironically, in the end, Subbulakshmi's father was paralysed at the end knowing that the password which was used by him to access his black money was changed. After that Subbulakshmi and Kanthaswamy were married and were living in northern India. Kanthaswamy saw some letters tied to a tree, just like the ones at the Lord Muruga temple. He reads one and puts it into his pocket with a secret smile, which meant he will pursue being the anthropomorphic rooster again.

Xavier (Romain Duris), a 24-year-old French student, leaves his country for the ERASMUS programme in Barcelona as part of his professional pursuits, despite it being against the wishes of his girlfriend Martine (Audrey Tautou). Xavier manages to find an apartment with other students from England, Belgium, Spain, Italy, Germany, and Denmark, creating an ambiance of chaos and culture shock. The roommates begin to develop a unique companionship as they struggle together through their cultural and linguistic challenges in their program. Martine pays Xavier a visit and returns disappointed when she realizes things are not the same. Meanwhile Xavier develops a romantic affair with the French doctor's wife, seducing her with tips he has been learning from his Belge lesbian roommate. The English roommate Wendy's brother William visits for some time and turns out to be quite abrasive with his culturally insensitive comments, creating tension among the roommates. Martine eventually breaks up with Xavier, bringing him to depression and hallucinations. When Xavier seeks the French doctor's advice, he reveals that Anne-Sophie told him everything and tells him to stop seeing her. As the discordance among the roommates escalates, their friendship is repaired as they team up to help Wendy elude a sticky situation (her boyfriend Alister makes a surprise visit while she was hooking up with an American). After leaving Barcelona and bidding farewell to his many close friends, Xavier returns to Paris and gets the job at the Ministry but realizes he misses his experiences that now make him a different person. He subsequently runs away on his first day and pursues his dream to become a writer, recounting the story of his experiences in the Auberge Espagnole.

An affair develops between them, but the possessive Angelo has her followed, and threatens her with dire consequences if she leaves him, mirroring Mario's own feelings and actions towards Laura before Elena entered his life.
