Gildersleeve's Ghost is a 1944 American comedy film directed by Gordon Douglas from an original screenplay by Robert E. Kent. It is the fourth and final film in the Gildersleeve's series, all of which were produced and distributed by RKO Radio Pictures, based on the popular NBC radio program, The Great Gildersleeve, created by Leonard L. Levinson. Released on September 6, 1944, the film stars Harold Peary, Marion Martin, Richard LeGrand, Amelita Ward, Freddie Mercer, and Margie Stewart.

The duo of Jerry Miles and Mike Strager are employed as Broadway press agents. Miles and Strager's latest idea is to hire a genuine zombie for the opening of a new nightclub. The boys head to the Caribbean island of San Sebastian where they meet the beautiful cabaret singer Jean la Dance. In exchange for her help, Jean wants passage off the island. Miles and Strager eventually meet up with the zombie expert Professor Renault. Unknown to them, the professor's zombie has captured Jean and brought her to the Professor's secret laboratory, While Miles and Strager investigate the house, Jean awakes to find herself gagged and strapped to a table as the Professor's next test subject. Before he can proceed to give Jean the serum his guard dogs detect intruders. Jean is quickly spirited to a secret dungeon where she is tied up but manages to escape. Strager becomes "zombified" by being under the spell of Renault's secret formula and Miles, la Dance, and Strager return home. When Strager comes out of his trance, the boys must face the wrath of Ace Miller, a nightclub owner, who is more frightening then anything they've seen yet.

The movie opens with Mystery, Inc. being pursued by a moat monster. After an accident with Scooby-Doo (Scott Innes), he is caught and revealed to be a counterfeiter. This is actually an old case of the now-dissolved Mystery, Inc., who went their separate ways because they were frustrated with their mysteries having all involved mere crooks in monster costumes, as told on a talk show program by Daphne Blake (Mary Kay Bergman), who now, along with Fred Jones (Frank Welker), is running a successful TV series Coast to Coast with Daphne Blake, which involves her and Fred going to historic and inspiring places all across the country. For the show's upcoming second season, Daphne intends to visit haunted and mysterious places and track down real ghosts. When Daphne confesses on air that she misses the gang and that she wishes they were with her, Fred arranges a reunion and contacts Velma Dinkley (B.J. Ward), who has gone on and become the proprietor of a mystery bookstore, and Scooby and his owner Shaggy Rogers (Billy West), who have been bouncing from job to job, including working in customs at an airport, from which they were just fired after eating all the confiscated imported foods. The reunited gang surprise Daphne on her birthday and they go for a trip to Louisiana for her show.

In an old Hollywood mansion, the spirit of a butler inhabits a grandfather clock.
