A decade after the events of the first film, Dr. Arthur Neuman is giving a tour of the hall of Norse mythology in Edge City Museum. When Dr. Neuman reaches the part concerning Loki's mask, a man in black becomes increasingly anxious. Dr. Neuman mentions that Loki created the mask and unleashed it on Earth, and that those who wear the mask would have the powers of Loki. When Dr. Neuman mentions that Odin punished Loki with imprisonment, the stranger becomes very angry and transforms, revealing himself to be Loki. The tourists panic and flee, but Dr. Neuman stays to argue with the angry god. Loki takes the mask, but realizes it is a fake. In anger, he removes Dr. Neuman's still talking face from his body and puts it on the mask stand, before getting rid of the guards and storming out of the museum in a whirlwind of rage.

Eight years after the events of the first film, Miami Police Department (MPD) narcotics division detectives Marcus Burnett and Mike Lowrey are investigating the flow of highly-potent ecstasy into the city. Their surveillance of boats coming in from Cuba leads them to a Ku Klux Klan meeting and drug drop in a swamp docks outside Miami. The subsequent raid on the Klan members proves to be a disaster; malfunctioning radios delay back-up, causing a firefight when Mike accidentally shoots Marcus in the buttocks, while the Klan members are revealed to be mere small-time buyers and not the distributors of the ecstasy. The incident leaves Marcus to further question if he still wants to partner with Mike, while Mike fears that Marcus may discover his new relationship with Marcus' sister, Syd.

Marcus Burnett (Martin Lawrence) and Mike Lowrey (Will Smith) are best friends and detectives in the narcotics division of the Miami Police Department. One night, $100 million of Mafia seized heroin is stolen from a secure police vault. This is a major blow to Burnett and Lowrey, because it was the biggest drug bust of their careers. Two months have passed since a mutated strain of mad cow disease mutated into "mad person disease" that became "mad zombie disease" which overran the entire United States population, turning American people into vicious zombies. Unaffected college student "Columbus" (Jesse Eisenberg) is making his way from his college dorm in Austin, Texas to Columbus, Ohio to see whether his parents are still alive. He encounters "Tallahassee" (Woody Harrelson), another survivor who is particularly violent in killing zombies. Though he does not appear to be sociable, Tallahassee reluctantly allows Columbus to travel with him. Tallahassee mentions he misses his "puppy" that was killed by zombies, as well as his affinity for Twinkies, which he actively tries to find. Survivors of the zombie epidemic have learned that it is best not to grow attached to other survivors, because they could die at any moment, so many have taken to using their city of origin as nicknames, i.e. "Columbus" is from Columbus, Ohio.
