To celebrate his upcoming marriage to Tracy Garner (Sasha Barrese), Doug Billings (Justin Bartha) travels to Las Vegas with his best friends Phil Wenneck (Bradley Cooper) and Stu Price (Ed Helms), as well as Tracy's brother and Doug's future brother-in-law Alan (Zach Galifianakis), in Doug's future father-in-law's vintage Mercedes-Benz. In 2010, three friends are miserable with their lives: Adam Yates (John Cusack) has been dumped by yet another girlfriend, and his geeky 20-year-old nephew Jacob (Clark Duke) lives in his basement, playing Second Life, with no idea who his father is. Lou Dorchen (Rob Corddry) is a party animal way past his prime. Two years after the events in Bangkok, Leslie Chow (Ken Jeong) escapes from a maximum security prison, using a riot as cover. A low-level marijuana dealer named David Clark (Jason Sudeikis) who lives in Denver is robbed of his money and stash, some of which he owes to his wealthy drug lord supplier Brad Gurdlinger (Ed Helms). After David is brought to him, Gurdlinger forces David to smuggle marijuana from Mexico as a solution to clear his debt. Realizing that one man attempting to get through customs is too suspicious, he hires a middle-aged stripper neighbor stage-named Rose (Jennifer Aniston), a runaway and petty thief named Casey (Emma Roberts), and his 18-year-old neighbor Kenny (Will Poulter), to pose as a bogus family called the "Millers". The film is about Melanie Porter (Raven-Symoné), a 17-year-old college-bound girl who is getting ready to graduate from high school and really wants to go to Georgetown University.
