The movie was directed by Michael Curtiz and produced by Arthur Schwartz, with Jack L. Warner as executive producer. The screenplay was written by Charles Hoffman, Leo Townsend and William Bowers.
The music score by Ray Heindorf and Max Steiner was nominated for an Academy Award. The film features several of the best-known Porter songs, including the title song, "Night and Day", "Begin the Beguine" and "My Heart Belongs to Daddy".
Alexis Smith plays Linda Lee Porter, Porter's wife of 35 years. Monty Woolley and Mary Martin appear as themselves, and the rest of the cast includes Jane Wyman, Eve Arden, Alan Hale, Dorothy Malone, Donald Woods, and Ginny Simms.
The film is a highly fictionalized and sanitized version of Cole Porter's life, leaving out amongst other things references to his homosexuality. A later film biography of Porter, the 2004 De-Lovely with Kevin Kline and Ashley Judd, dealt more frankly (though some said not enough) with his sexuality and includes a scene where Kline and Judd as Cole and Linda Lee watch Night and Day before its release and comment on its lack of realism and "happy ending".

In the early days of World War II, Cohan comes out of retirement to star as President Roosevelt in the Rodgers and Hart musical I'd Rather Be Right. On the first night, he is summoned to meet the President at the White House, who presents him with a Congressional Gold Medal (in fact, this happened several years previously). Cohan is overcome and chats with Roosevelt, recalling his early days on the stage. The film flashes back to his supposed birth on July 4, whilst his father is performing on the vaudeville stage.

Elvira Kent (Janis Paige) and her husband Michael (Don DeFore) suspect each other of cheating. The film opens in Los Angeles, where Doug Blake (Carson) is dumped as a manager by Gary Mitchell (Bowman). Frank Fay, as a Mexican named Don Carlos, rides into a small Texas border settlement on the Fourth of July in the early 1880s. He is accompanied by his two inseparable companions, played by Georgie Stone and George Cooper. The day is being celebrated in the style of a Spanish fiesta. Fay challenges a rough Texan, played by Noah Beery, to a duel, only to find himself invited to undertake the dangerous task of capturing a cattle rustler who has been stealing cattle from the Lazy Y Ranch. He accepts the task on the promise of receiving seven thousand dollars in gold if he can return both the thief and the stolen cattle within ten days. During the next nine days, Fay spends his time making love to every pretty girl he meets, serenading many of theme by singing the theme song to the film while playing his guitar, while his two companions join in the harmonizing. He lies to them all, telling each girl exactly what she wishes to hear. Throughout all this time he does nothing towards earning his reward. On the tenth day, he captures the cattle rustler and turns up the cattle to everyone's surprise by using a simple method that no one had thought of. He rides back to Mexico with his latest conquest in his arms.
