The story begins with a military truck transporting barrels of Trioxin. The soldier, driving the vehicle through a downpour, is unaware when a barrel breaks loose and falls into a river. The next morning, a young boy, Jesse Wilson, is at the cemetery with 2 local bullies. The trio investigate the Trioxin tank that they find, and Jesse warns them that they should not tamper with it. The bullies trap Jesse in a derelict mausoleum and leave him. They then return to the trioxin tank and manage to release the toxic gas. A van pulls up to the graveyard, introducing the characters Ed, Joey, and Brenda. Ed explains to Joey that they are there to rob graves; Brenda expresses her fears for cemeteries, but Joey assures her that it will be worth their time and leaves Brenda in the van. He heads into the cemetery with Ed. They decide to loot the mausoleum and open the locked doors, releasing Jesse, who immediately runs home. At his home, Jesse watches his older sister, Lucy, doing aerobic exercises to a work-out video; she tells Jesse to do his homework or he will be grounded.

At a medical supply warehouse in Louisville, Kentucky, a foreman named Frank tries to impress the company's newest employee, Freddy, by showing him military drums that accidentally wound up in the basement of the building. The drum contains the remains of an army experiment gone wrong that inspired the film Night of the Living Dead. Frank accidentally unleashes the toxic gas inside the barrel, knocking them unconscious. When they wake up, Frank and Freddy discover that the body inside the tank has disappeared, believing it to have probably melted. The gas reanimates a corpse inside a meat locker, forcing Frank and Freddy to call upon their boss and warehouse owner Burt Wilson to help them deal with the situation. When the three fail to kill the walking cadaver by damaging its brain and decapitating it, Burt decides to bring the zombie to the nearby mortuary to have its dismembered parts burned in a gambit to destroy it once and for all.

About eight years after a zombie apocalypse, R, a zombie, spends his days wandering around an airport which is now filled with hordes of his fellow undead, including his best friend M. Shaun (Simon Pegg) is a 29-year-old electronics shop salesman with no direction in life. By the year 2056, an epidemic of organ failures has devastated the planet. The megacorporation GeneCo provides organ transplants on a payment plan. Clients who default on payments are hunted down by Repo Men: skilled assassins contracted by GeneCo to repossess organs, usually killing the clients in the process. The CEO of GeneCo, Rottissimo "Rotti" Largo (a listed in a newspaper article about his kids), discovers he is terminally ill. Rotti's three children, Luigi Largo, Pavi Largo, and Amber Sweet, who changed her last name as a stage stunt to replace a popular singer she's jealous of, bicker over who will inherit GeneCo. Rotti believes none of his children are worthy heirs, as they consistently embarrass him with their robust attitudes, and instead plans to pass on his fortune to Shilo, the daughter of his ex-fiance Marni.

This movie opens with a news montage explaining that it is set in a dystopic near-future in which George W. Bush has been elected to a fourth term. The United States Congress has been disbanded; public nudity is banned; the United States is embroiled in wars with France, Iraq, Afghanistan, Iran, Pakistan, Syria, Venezuela, Canada, and Alaska. With more wars than there are soldiers to fight them, a secret laboratory run by Dr. Chushfeld (Brad Milne) in fictional Sartre, Nebraska, has developed a virus to re-animate dead Marines and send them back into battle. However, this virus has broken containment and infected test subjects and scientists, and they are at risk of escaping the lab. A team of Marines codenamed the "Z" Squad is sent in to destroy the zombies. One of the Marines named Byrdflough (Zak Kilberg) is bitten but escapes. He ends up in an alley outside an underground strip club named "Rhino". The Marine dies and awakens as a zombie who goes into the strip club.
