In a world populated by anthropomorphic vehicles, the last race of the Piston Cup championship ends in a three-way tie between retiring veteran Strip "The King" Weathers, infamous runner-up Chick Hicks, and rookie Lightning McQueen. The tiebreaker race is scheduled for one week later at the (fictional) Los Angeles International Speedway in California. Lightning is desperate to win the race, since it would allow him to leave the unglamorous sponsorship of Rust-Eze, a rust treatment for old cars, and allow him to take The King's place as the sponsored car of the lucrative Dinoco team. Eager to start practice in California as soon as possible, he pushes his big rig, Mack, to travel all night long. While McQueen is sleeping, the exhausted Mack drifts off and is startled by a gang of four reckless street racers, causing McQueen to fall out the back of the trailer and into the road. McQueen wakes in the middle of traffic and speeds off the highway to find Mack, only to end up in the run-down desert town of Radiator Springs, and inadvertently ruining the pavement of its main road.

The film begins with GloboTech Industries acquiring the Heartland Toy Company. CEO Gil Mars tells remaining toy designers Larry Benson and Irwin Wayfair to develop actual live-action toys capable of "playing back". Mars selects Larry's action figures, the Commando Elite, for the project and Irwin's educational toys, the Gorgonites, for their enemies. After Mars sees a commercial showing the toys doing activities they are incapable of doing, he accuses them of false advertising, saying Globotech delivers on what it promises. Mars then orders Benson to have the toys on the shelves in three months acting just as they do in the commercial or get fired. Faced with such a tight deadline, Benson forgoes safety testing, then uses Irwin's password and chooses GloboTech's overly powerful, X1000 intelligent AI munitions microprocessor integrated circuit to control the toys.

On the streets of Fifth Avenue, an orphaned kitten named Oliver is left abandoned after his fellow orphaned kittens are adopted by passersby. Wandering the streets by himself in search of someone to adopt him, Oliver meets a laid-back dog named Dodger who assists the kitten in stealing food from a hot dog vendor named Louie. Dodger then flees the scene without sharing his bounty with Oliver. Oliver follows Dodger all throughout the streets until he eventually arrives at the barge of his owner, a pickpocket named Fagin, along with his meal, to give to his friends: Tito the Chihuahua, Einstein the Great Dane, Rita the Saluki and Francis the Bulldog. Oliver sneaks into their home, located below the city's docks, and is discovered by the dogs. After a moment of confusion, Oliver is then received with a warm welcome. Fagin, owner of the dogs, comes in and explains that he is running out of time to repay the money he borrowed from Sykes, a ruthless shipyard agent and loan shark. Sykes tells Fagin the money must be paid in three days, or else. Sykes' Dobermans Roscoe and DeSoto attack Oliver but the cat is defended by Fagin's dogs. Immediately thereafter, a depressed Fagin returns to the barge, lamenting that he has only three days to find the money he owes Sykes. After the dogs cheer him up, Fagin is introduced to Oliver, and, considering that they all need help, accepts him into the gang.

A young, naive boy named Billy Forrester (Luke Benward) has a weak stomach and vomits easily. He and his parents, Mitch (Tom Cavanagh), Helen (Kimberly Williams-Paisley), and his little brother, Woody (Ty Panitz), have just moved to a new town. Billy tells his mother that he doesn't want to go to school because he will be "the new kid". She assures him that he will make friends and everything will be okay. Tadeo Jones (voiced in English by Kerry Shale) is a bricklayer from Spain who lives in Chicago, working on the construction of the city's subway. Ever since he was five years, he has dreamed of being a professional archaeologist. After being fired from his job as a bricklayer for daydreaming and for his dog urinating on the construction foreman, Tad visits his friend Professor Humbert (voiced by Mac McDonald), asking for the professor's help in investigating an apparently vintage bottle of Coca-Cola Tad found on the construction site. The professor receives a letter from his friend Professor Lavrof summoning him to Peru to re-unite his half of a stone "key" that will unlock the legendary Incan city of Paititi. Arriving at the airport, Professor Humbert suffers an accident after taking the wrong pills and is taken to the hospital. Meanwhile, a member of the nefarious firm Odysseus Inc. is spying on them and sends the photo to the other members who are in the Sechura Desert.

Five years after Agent K's retirement, Agent J (Will Smith) is now a top operative for the MIB, the New York City-based agency that secretly monitors and regulates extraterrestrials' activity on Earth.
