On the day of her wedding to news weatherman Derek Dietl, Susan Murphy is struck by a meteorite and its energy causes her to suddenly grow over fifty feet in height during the ceremony. She is tranquilized and awakens in a top secret government facility that houses monsters without human knowledge. She meets the warden W.R. Monger, and fellow inmates: Dr. Cockroach P.H.D., a mad scientist who became half cockroach after an experiment; B.O.B. Shrek and Fiona return from their honeymoon to find they have been invited by Fiona's parents to a royal ball to celebrate their marriage, an event Shrek is reluctant to participate in. Many days after bidding the penguins goodbye, Alex has a nightmare about himself and his friends still being stranded in Africa and finding that they have all gotten old. Seth (Jonah Hill) and Evan (Michael Cera) are two high school seniors who lament their virginity and poor social standing. Best friends since childhood, the two are about to go off to different colleges, as Seth didn't get accepted into Dartmouth like Evan. After Seth is paired with Jules (Emma Stone) during Home-Ec class, she invites him to a party at her house later that night. Later, Fogell (Christopher Charles Mintz-Plasse) comes up to the two and reveals his plans to obtain a fake ID during lunch. Seth uses this to his advantage and promises to bring alcohol to Jules' party. Fogell's fake ID is met with derision by Seth and Evan, as it states that Fogell's name is simply "McLovin". After contemplating their options, Seth decides they have no choice but to have Fogell buy the alcohol with his fake ID. Fogell goes in and successfully buys the alcohol, but is interrupted when a robber enters the store, punches him in the face, and takes money from the cash register.

One summer on the Island of Sodor, Spencer (voiced by UK: Matt Wilkinson/US: Glenn Wrage) arrives to help the Duke (voiced by UK: Matt Wilkinson/US: William Hope) and Duchess (voiced by UK: Teresa Gallagher/US: Jules de Jongh) with the construction of their new summer house. After being an absolute nuisance to all of the other engines by teasing and bossing them around, Thomas (voiced by UK: Ben Small/US: Martin Sherman) accepts Spencer's challenge to a contest of strength, taking heavy cargo around the island. Thomas's brakes fail after climbing a tall hill, and he runs out of control through an overgrown, abandoned line. There, Thomas finds an old abandoned Japanese engine named Hiro (voiced by Togo Igawa). Hiro doesn't want the Fat Controller (voiced by UK: Keith Wickham/US: Kerry Shale) to find out, fearing he will be scrapped, so Thomas promises to keep him his secret and work to make Hiro "Master of the Railway" once again. Later, he's taken to the Sodor Steamworks for repairs, where he finds old parts for Hiro. He takes them with permission from Victor (voiced by UK: Matt Wilkinson/US: David Bedella), the Steamworks engine, only to discover that Hiro's hiding place is dangerously close to the Duke and Duchess's new summer house.

Alison Scott (Katherine Heigl) is a career-minded woman who has just been given an on-air role with E! and is living in the pool house with her sister Debbie's (Leslie Mann) family. Ben Stone (Seth Rogen) is laid-back and sardonic. He lives off funds received in compensation for an injury and sporadically works on a celebrity porn website with his roommates (Jay Baruchel, Jonah Hill, Jason Segel and Martin Starr), in between smoking marijuana or going off with them at theme parks such as Knott's Berry Farm. While celebrating her promotion, Alison meets Ben at a local nightclub. After a night of drinking, they end up having sex. Due to a misunderstanding, they don't use protection: Alison uses the phrase "Just do it already" to encourage Ben to put the condom on, but he misinterprets this to mean to dispense with using one. The following morning, they quickly learn over breakfast that they have little in common and go their separate ways, which leaves Ben visibly upset.
