Shortly after the dawn of the new millennium, during construction on the London Underground, workers penetrate an underground cave. A huge dragon emerges from hibernation, incinerating the workers with its flaming breath. The only survivor is a boy, Quinn Abercromby (Ben Thornton), whose mother Karen (Alice Krige) is the construction crew chief; Karen is crushed to death while protecting Quinn as the dragon climbs to the surface. It flies out of the Underground and more dragons appear, multiplying rapidly. Shown in newspaper clippings and a voiceover, scientists discover that dragons are a lost species responsible for the extinction of the dinosaurs by incinerating them and eating their ash, as well as ice ages. The dragons are speculated to hibernate after destroying most of the Earth's creatures until the planet repopulates. After the dragons' awakening humanity resists militarily, including a 2010 use of nuclear weapons; this, however, hastens the destruction and within a few years humans are nearly extinct.

In 2013, Earth's coastal cities come under attack from Kaijus, colossal beasts that periodically emerge from a dimensional portal on the Pacific Ocean floor called "The Breach." To fight them, the Pacific Rim nations build the Jaegers, equally colossal humanoid war machines. Each Jaeger is controlled by two or more pilots whose brains are linked, called "drifting" to share the heavy mental load of operating the machine. The Jaegers are initially effective, but many are destroyed as the Kaijus grow more powerful and their attacks become more frequent. In 2025, the participating governments deem the Jaeger program unsustainable and discontinue it in favor of building defensive coastal walls. The four remaining Jaegers are redeployed to Hong Kong to protect the coast until the walls are completed. Jaeger commander Stacker Pentecost resolves to make a last-ditch attempt to end the Kaiju threat by destroying The Breach with a nuclear weapon, even though past nuclear strikes on the portal have been unsuccessful.

Charlotte, a young woman, is abducted by Baron Meier Link, a vampire nobleman who is known not to harm humans needlessly. Charlotte's father hires D, a dhampir, to find her and kill her humanely if she turns into a vampire. At the same time, her older brother also hires the notorious Marcus brothers for backup. Among them is a woman named Leila, who hunts vampires because of a personal grudge rather than for monetary gain. The two parties (D and the Marcus brothers) race inexorably after Meier Link. While sailing, Sinbad (John Phillip Law) comes across a golden tablet dropped by a mysterious flying creature. He wears the tablet as an amulet around his neck. A group of vampires look for the hidden resting place of Drake, also known as Dracula—the genetic "father" of their species. Darren Shan and his best friend Steve Leonard visit a traveling freak show. Steve believes a performer, Larten Crepsley, is a vampire. Steve talks to Crepsley after the show and asks to become a vampire. Crepsley initially refuses but is talked into testing Steve's blood, that he spits out telling Steve he has bad blood. King Kong, after being shot down from the World Trade Center, is kept alive in a coma for about 10 years at the Atlanta Institute, under the care of surgeon Dr. Amy Franklin (Linda Hamilton). In order to save Kong's life, Dr. Franklin must perform a heart transplant and give Kong a computer-monitored artificial heart. However, he lost so much blood that a transfusion is badly needed, and to complicate matters, Franklin says there no species of ape or other animal whose blood type matches Kong's. Enter adventurer Hank "Mitch" Mitchell (Brian Kerwin), who captures a giant female gorilla, who is dubbed "Lady Kong"; in Borneo (Mitchell theorizes that Borneo and the island from the first movie were once part of the same landmass), bringing her to the Institute so her blood can be used for Kong's operation. The transfusion and the heart transplant are a success, but Kong escapes along with the female.

The crew of the space shuttle Churchill finds a 150-mile long spaceship hidden in the corona of Halley's Comet. The crew finds hundreds of dead, shrivelled bat-like creatures and three naked humanoid bodies (two male and one female) in suspended animation within glass containers. The crew recovers the three aliens and begins the return trip to Earth.
