The dim-witted but good-natured Derek Zoolander is ousted as the top male fashion model by the rising star, Hansel, and his reputation is further tarnished by a critical article from journalist Matilda Jeffries. High school student Dan Bartlett (John Cusack) misses the plane he was supposed to be on with his rich girlfriend and her family on the way to a Caribbean vacation during a school break. He flies there alone, and runs into a series of characters and misadventures as he tries to catch up. Charlie Sykes and Rachel Clark are a young all-American couple that decide to run away and elope in Las Vegas. On the road to Vegas, they ignore the warning of a local gas station attendant named Sam. They take an abandoned backroad where Rachel is kidnapped by a zombie Hell Cop who takes her to hell. Charlie goes back to Sam, and Sam explains what the Hellcop is and how to save her. Sam then gives Charlie a shotgun with special ammo and a car that holds a special attribute. On the highway, Charlie meets other dead people that live in Hell and even a motorcycle gang. On the road, he meets a mechanic named Beezle, with his young apprentice. Beezle gives him tips on how to save his girlfriend and even brings him back to life. After Charlie rescues Rachel, Beezle reveals himself to be Satan and proposes a deal to let them, and his apprentice, go free if they can defeat the Hell Cop in a race to the portal that connects Earth and the backroads of Hell.

In the metropolis of Champion City, the would-be superhero team of Mr. Furious (Ben Stiller), The Shoveler (William H. Macy), and The Blue Raja (Hank Azaria) attempt to make a name for themselves, but their suspect skills make them ineffective, and they find themselves upstaged by the city's most successful superhero, Captain Amazing (Greg Kinnear). However, Amazing is finding that his effectiveness at fighting crime has practically made his job obsolete, and without any worthy adversaries remaining (some of them are either dead, in exile, or still in jail), his corporate sponsors are beginning to pull their funding. To create a need for himself, Amazing uses his alter ego, billionaire lawyer Lance Hunt, to argue for the release of insane supervillain Casanova Frankenstein (Geoffrey Rush). The plan backfires; once reunited with Tony P (Eddie Izzard) and his Disco Boys, Casanova Frankenstein blows up the insane asylum, captures Amazing, and prepares to unleash a doomsday device: the "Psycho-frakulator", which lethally bends reality. Four friends who recently graduated from college live together in Houston, Texas.
