In 932, King Arthur, along with his squire, Patsy, recruits his Knights of the Round Table: Sir Bedevere the Wise, Sir Lancelot the Brave, Sir Galahad the Pure, Sir Robin the Not-Quite-So-Brave-As-Sir-Lancelot, and the aptly named Sir Not-appearing-in-this-film. On the way, Arthur battles the Black Knight who refuses to surrender heedless of his arms and legs being severed. The knights reach Camelot, but following a song-and-dance cutaway, Arthur decides not to enter, because "'tis a silly place". They are instructed by God to seek the Holy Grail.

The film begins with a stand-alone 17-minute supporting feature entitled The Crimson Permanent Assurance (directed by Gilliam). A group of elderly office clerks in a small accounting firm rebel against their emotionlessly efficient, yuppie corporate masters. They commandeer their building, turn it into a pirate ship, and sail into a large financial district, where they raid and overthrow a large multinational corporation (before ultimately sailing to the edge of the earth and falling off).

The film begins with an animated version of director Terry Gilliam reading from a children's storybook. The story, "Don the Cockroach", opens with a palace high atop a mountain. Inside the wall of one of its rooms lives a cheerful cockroach named Don, who is shown frolicking and generally enjoying his life as a cockroach. One day, as Don scurries across a kitchen floor, he is suddenly crushed by a foot. The narrator Gilliam remarks that this does not matter, as cockroaches are not very interesting. The story then shifts to the man who crushed Don, then to his brother, then to the brother's friend, then to the friend's employer, and so forth. Eventually the chain of interconnected people reaches a cleaning lady who is involved in a search-and-destroy mission against a colony of cockroaches, one of whom is named Don. The film then loops back to Don, replaying some of the earlier events until a gruff voice interrupts the narrator with "You already said that!" and the sound of a punch. An intertitle appears, announcing that the animator responsible for the previous cartoon has been sacked, and promising that the next one will be better.

The film begins in an unnamed war-torn European city in the late 18th century (dubbed "The Age of Reason" in an opening caption), where, amid explosions and gunfire from a large Turkish army outside the city gates, a fanciful touring stage production of Baron Munchausen's life and adventures is taking place. Wilhelm Grimm (Matt Damon) and Jakob Grimm (Heath Ledger) arrive in French-occupied Germany during the early 1800s. They go to Karlstadt to rid the town of a witch's ghost. After killing the "ghost", it is revealed that the Brothers Grimm have actually set up a fake witch to trick the town. Afterwards, as they are celebrating, Italian torturer Mercurio Cavaldi (Peter Stormare) takes them to the French General Delatombe (Jonathan Pryce). Delatombe forces them to solve a mystery: The girls of the small village of Marbaden are disappearing, and the villagers are convinced that supernatural beings are responsible. The Grimms are charged with finding who is responsible, and they soon discover that it is the work of a real supernatural force: a beautiful, yet evil, 500-year-old Thuringian Queen (Monica Bellucci) stealing young girls to restore her own beauty. Will and Jake have a complicated relationship; Jake is the smaller, younger, more sensitive one that Will feels he needs to protect. Will is often very hard on Jake (dating all the way back to their childhood, when Jake spent their money that was to be used for medicine for their dying sister on "magic beans") and orders him around. Will is somewhat of a womanizer and wants to make money, whereas Jake is more interested in fairy tales and adventures. Jake feels that Will doesn't care about or believe in him; but Will is just frustrated about the way Jake acts so spontaneously, making it hard for Will to protect him.

Jack Lucas (Bridges), a selfish, misanthropic shock jock, becomes suicidally despondent after his insensitive on-air comments inadvertently prompt an unstable caller to commit a mass murder-suicide at a popular Manhattan restaurant. Three years later, Jack is working with his girlfriend Anne (Ruehl) in a video store in a mostly drunken, depressed state. One night while on a bender, he attempts suicide. Before he can do so, he is mistaken for a homeless person and is attacked and nearly set on fire by thugs. He is rescued by Parry (Williams), a deluded homeless man who is on a mission to find the Holy Grail, and tries to convince Jack to help him. Jack is initially reluctant, but comes to feel responsible for Parry when he learns that the man's condition is a result of witnessing his wife's horrific murder at the hands of Jack's psychotic caller. Parry is also continually haunted by a hallucinatory red knight, who terrifies him.
