The film follows Dr. Hess Green (Duane Jones), a wealthy black anthropologist who is doing research on the Myrthians, an ancient African nation of blood drinkers. One night, while staying in Green's lavish mansion --richly decorated with African art--, Green's unstable assistant George Meda (writer/director Gunn), attempts suicide. Green successfully talks him down, but later that night Meda attacks and stabs Green with a Myrthian ceremonial dagger, and then kills himself. Green, discovering the body, drinks Meda's blood and becomes a vampire endowed with immortality and a need for fresh blood. Though he steals several bags of blood from a doctor's office, he quickly finds that he needs fresh victims.

A year later in 1906, following the events of the previous film, Dracula has been destroyed. Monsignor Ernest Mueller (Rupert Davies) comes to the village on a routine visit only to find the altar boy is now a frightened mute and the priest (Ewan Hooper) has lost his faith. The villagers refuse to attend Mass at the Catholic church because the shadow of Dracula's castle touches it. To bring to an end the villagers' fears, Mueller climbs to the castle to exorcise it.

Renfield is a solicitor traveling to Count Dracula's castle in Transylvania on a business matter. The people in the local village fear that vampires inhabit the castle and warn Renfield not to go there. Renfield refuses to stay at the inn and asks his carriage driver to take him to the Borgo Pass. Renfield is driven to the castle by Dracula's coach, with Dracula disguised as the driver. En route, Renfield sticks his head out the window to ask the driver to slow down, but sees the driver has disappeared; a bat leads the horses.

A Miami businessman, John Stone, receives a parcel from England containing two old bottles of Slivovitz brandy from his recently deceased ancestor, and after drinking both bottles, becomes a vampire. Anna Quarrels (Rhonda Griffin), librarian in the Rare Books Room, is approached by Mr. Jamison from the University of Chicago who wishes to study Mary Shelley's original manuscript for "Frankenstein; or, The Modern Prometheus". After he's finished, Anna is about to return the manuscript to the stacks when she discovers that Jamison has switched blank paper for the manuscript and walked out of the library with it. Investigating, she learns he used a fake I.D. to access the Rare Books Room and so hires Private Detective, David Raleigh (Justin Lauer) to track him down. David finds fingerprints on the sign-in sheet and discovers the man claiming to be Jamison is really Dr. Winston Berber, an unscrupulous scholar with doctorates in Physics, Mathematics, Folklore, and Philosophy. Berber (Bill Moynihan) is meanwhile gloating over his collection of rare manuscripts; along with the Shelley manuscript, he has obtained the originals of Guy Endore's "The Werewolf of Paris" (1933) and James Putnam's "Mummy" (1993). He now seeks the first edition of Bram Stoker's "Dracula" (1897) to complete his collection. Berber has invented an "Archetype Inducer" and plans to use the manuscripts to bring the four greatest monsters from horror history to life to do his bidding.
