It is 1940. When the movie begins, film star Viviane Denvert sits in the audience of a premiere of her new movie and notices a man who keeps staring at her. She is disturbed, and when the film is over and the audience has finished praising her, she rushes home, discovering that she is pursued by that same man. He chases her into her apartment.
Forty-year-old Guy Trilby discovers a loophole in the Golden Quill Spelling Bee which stipulates that participants must have not graduated from the eighth grade, allowing him to enter since he dropped out of middle school. He attends and wins a regional spelling bee, and progresses to the national competition after confrontations with both the parents of children and the spelling bee hosts. He is accompanied by Jenny Widgeon, a relatively unknown journalist hoping to make a story out of his participation in the bee. On the flight to the national spelling bee, Guy meets Chaitanya Chopra, a 10-year-old entrant in the bee who persistently attempts to befriend Guy. Upon his arrival in Los Angeles, Guy meets the director of the spelling bee, Dr. Bernice Deagan, who expresses anger at his participation in the bee and places him and his reporter in a cheap hotel. Guy learns that Chaitanya is staying in the same hotel, and takes him out one night to expose Chaitanya to the wilder side of life: stealing food, drinking, and briefly hiring a prostitute.
The eponymous theatre troupe is rehearsing the title song with hopes of finding success through being picked up for a syndicated television show. Heidi, the star of the show, is insulted by pornographic director Trevor and complains to her boss and lover, Bletch, who is actually in an adulterous relationship with Samantha. Meanwhile, Robert, the new member of the team, arrives at the theatre and immediately falls in love with another newcomer, Lucille. Samantha confronts Heidi, insults her, and reveals her relationship with Bletch. Robert confesses his love to Lucille, and the two become engaged. Sidney, Arthur's friend and member of the team, receives a visit from his ex-wife Sandy with his alleged son Seymour (who comically very strongly resembles Sidney). Sandy informs him she will be preparing a paternity case against him.
Chris (Steve Oram) is a caravan fan and aspiring writer who takes his girlfriend Tina (Alice Lowe) on a road trip, much to the chagrin of Tina's mother (Eileen Davies), who has never forgiven Tina for the death of their dog "Poppy". At their first stop, a tramway museum (filmed at Crich Tramway Village, the home of the National Tramway Museum), Chris confronts a man (Tony Way) who is littering, and the man refuses to pick up his rubbish. When they get back to their car, Chris runs him over and kills him, which upsets the couple but they continue onward on their trip. Chris claims that the death was an accident, but his smirk—visible to the camera but not to Tina—in the immediate aftermath makes it clear it was deliberate.

In the midst of preparing for his wedding and desperate for a break from his "Bridezilla" fiancée Laura (Cameron Diaz), Kyle Fisher (Jon Favreau) organizes a bachelor party in a Las Vegas hotel with his best friends: Charles Moore (Leland Orser), Robert Boyd (Christian Slater), and brothers Adam and Michael Berkow (Daniel Stern and Jeremy Piven), spending the night with drinks, drugs and a stripper/prostitute, Tina (Kobe Tai). Things go awry when Michael gets drunk and pays Tina extra money for sex in the bathroom only to accidentally slam her against a wall and impale a towel hook through her head, killing her instantly. As the group argues over what to do -- Adam insisting that they call the police, Boyd adamant about burying Tina's body in the desert -- a security guard comes to investigate the noise and sees Tina's corpse in the bathroom. In desperation, Boyd stabs the guard to death and the group is forced to dismember the bodies before taking them to the desert for burial.
Jerry is an upbeat man who works at a bathtub factory, and lives in a modified apartment above a bowling alley with his dog Bosco and his cat, Mr. Whiskers. Jerry is a man with an innocent, almost child-like demeanor, and suffers from delusions and hallucinations that manifest in the form of his pets talking to him. Bosco often represents his good intentions while Mr. Whiskers represents his more violent nature. One day, his manager compliments his hard work and chooses him to help organize an employee barbecue, and he gladly accepts the opportunity to work with his workplace crush, a British woman named Fiona. The following day he asks her out on a date. She initially agrees, though with reluctance, but then stands him up to go to a karaoke party with two other girls who work with her in the accounting department at work, Lisa and Alison. After the party, Fiona's car won't start, leading her to flag down Jerry as he drives by. Fiona offers to take him out for a late dinner to make up for standing him up on the original date, but on the way, Jerry accidentally hits a stag which crashes through his windshield. Jerry's hallucinations show the deer crying out in pain and begging Jerry to kill it so he slits the deer's throat. Fiona, terrified, runs off into the woods. Jerry pursues her and accidentally stabs her. Apologizing for his actions, Jerry kills Fiona to end her pain.

The film opens in Edinburgh. Narration by Angus the Hangman explains how the corpses of the hanged are transported to Dr. Robert Knox for dissection. Knox's rival, Dr. Alexander Monro, wants the steady supply of cadavers but is forced to rely on severed limbs for dissection. Monro's assistant Charles Darwin arrives with a forged letter directing that all corpses thenceforth must be sent to Monro. Angus tells Knox's assistant, Patterson, the news. Patterson delivers the message to Knox.
