In 2005, bookish student Morton Schmidt (Jonah Hill) and popular underachieving jock Greg Jenko (Channing Tatum) miss their school prom. Seven years later, the two meet again at the Police Academy and become firm friends and partners on bicycle patrol. They catch a break when they arrest Domingo (DeRay Davis), the leader of a one-percenter motorcycle gang, but are forced to release him after they failed to read him his Miranda rights.
Two years following their success in the 21 Jump Street program, Schmidt (Jonah Hill) and Jenko (Channing Tatum) are back on the streets chasing narcotics. However, after failing in the pursuit of a group of drug dealers led by Ghost (Peter Stormare), Deputy Chief Hardy (Nick Offerman) puts the duo back on the program to work for Captain Dickson (Ice Cube) – now located across the street at 22 Jump Street. Their assignment is to go undercover as college students and locate the supplier of a drug known as "WHYPHY" (Work Hard, Yes, Play Hard, Yes) that killed a student photographed buying it on campus.
Convicted thief Albert Ganz is working as part of a road gang in California, when a big Native American man named Billy Bear drives up in a pickup truck and asks for water to cool off his truck’s overheating radiator. Ganz and Billy exchange insults and proceed to stage a fight with each other, wrestling in a river, and when the guards try to break up the fight, Billy slips a gun to Ganz, and Billy and Ganz kill two of the three guards and flee the scene. Two days later, Ganz and Billy kill Henry Wong (John Hauk), who was one of their partners.
San Francisco cop Jack Cates is at the Hunter's Point Raceway checking out a known associate of the Iceman, Tyrone Burroughs, who for unknown reasons is giving some money to mechanic Arthur Brock.
The film is set in the 1950s in a large country residence as a family and its servants are preparing for Christmas. When the famous Pink Panther diamond is stolen again from Lugash, Chief Inspector Clouseau (Peter Sellers) is called on the case despite protests by Chief Inspector Dreyfus (Herbert Lom). While on the case, Clouseau is pursued by the Mafia. Clouseau first goes to London to interrogate Sir Charles Lytton (having forgotten that he lives in the South of France). Traveling to the airport, he accidentally blows up his car, but mistakenly believes it an assassination attempt, and disguises himself in a heavy cast on the flight, which causes complications in the air and on land and leads to an awkward introduction to the Scotland Yard detectives at Heathrow. Meanwhile, Dreyfus learns from Scotland Yard that Libyan terrorists have marked Clouseau for assassination; but permits him to continue.
