Gaylord "Greg" Focker (Stiller) is preparing to celebrate his twins' fifth birthday party. Things seem to go awry when Greg's father-in-law Jack Byrnes (De Niro) visits. Recently, Jack has been diagnosed with a heart condition and become embittered by his daughter Debbie's divorce from her husband, Bob (whose marriage was the social event in the first film and how Jack and Greg met), for cheating on her with a nurse. Jack's plan was originally to name Bob as his successor as head of the Byrnes family, but decides to pass the role to Greg, naming him "The Godfocker." Despite Greg reluctantly accepting the role, Jack begins to suspect Greg of infidelity when he sees him with a drug representative, Andi Garcia (Alba), who openly flirts with him, and the presence of Sustengo erection pills in Greg's house, which prompts Jack to think Greg is no longer sexually attracted to his wife, Pam (Polo). Furthermore, Jack starts to doubt Greg's ability to provide for his family when he appears reluctant to send his children to a private school.
Christopher and Leslie are a couple in their mid to late 20s who have no-strings-attached sex on a regular basis. The film tells the story of a young man named Anand (Salman Khan), who was deeply in love with a girl named Maya (Rimi Sen). He intentionally threw her into a swimming pool, thinking she could swim, for the sake of getting back at her pranks. However, Maya drowned. After her death, Anand's life is completely shattered and he becomes insane.
Preeti Vyas (Aishwarya Rai) is a young, vivacious woman. She comes forward as a witness to heinous assault carried by Bhavani Choudhry (Mukesh Rishi) and his men on a person who owes them money. The injured man is helped to the hospital by Preeti and a courteous and brave man called Avinash (Anil Kapoor), who holds strong morals and values. Preeti's testimony angers the Choudhry family and, as a result, Choudhry's younger brother, Babloo Choudhry (Puru Raajkumar), rapes her. Subsequently, she becomes a disgrace to her own family. Disowned by her family and secluded by society, she leaves her house with nowhere to go.
The film takes place over a summer by the seaside and follows three girls through a summer that will change their lives. The story is told by Keira St. George (Caroline Dhavernas), a girl who is trying to decide what to do with her life. She constantly throws away her college letter without reading it, but her father always retrieves it, annoying her more. Her two best friends are Glory Lorraine (Amanda Walsh) a beautiful but somewhat bitchy girl obsessed with marriage, and Lisa MacDougall (Holly Lewis), an awkward, born-again, religious girl who is obsessed with sex.
Alex (Gael Garcia Bernal) and Nica (Hani Furstenberg) are in love and engaged to be married. They are seasoned travelers on a trip in the country of Georgia, formerly a republic of the Soviet Union. They hire a local guide (Bidzina Gujabidze) to take them backpacking through the breathtaking scenery of the Caucasus Mountains.
Mohit Chadha (Ayushmann Khurrana) learns how to drive a car on the morning of his promotion from Junior Executive in Marketing to Senior Executive. He goes for lunch with Mayera (Sonam Kapoor) after the promotion, where he proposes to her. Meanwhile, Mayera's father, V.K. Sehgal (Rishi Kapoor), is transferred to Animal Husbandry. V.K. Sehgal does not approve of their wedding, as he thinks Mohit would not be able to provide for her and give her happiness. V.K. goes to Mohit's office and challenges him: if he passes a test, then he can marry Mayera. He calls Mohit to the club everyday to play squash. V.K. gets a retirement gift from all his staff.
Child psychologist Samuel Faulkner (Hugh Grant) has an ideal romance with ballet teacher Rebecca Taylor (Julianne Moore). Rebecca is thinking about marriage and children. Samuel is against the idea of marriage as he is happy with how things are between them. This all changes when Rebecca declares she is pregnant and when questioned by Samuel about her birth control she replies birth control is only 97% time effective. His fears mount due to his encounters with overbearing couple Marty (Tom Arnold) & Gail (Joan Cusack) and their unruly daughters, as well as the confusing advice he gets from his perpetually single artist friend (Jeff Goldblum). They meet their Doctor Kosevich (Robin Williams) who happens to be Russian. Samuel is confused and unsure about what to do. Feeling neglected Rebecca leaves him and moves in with Marty and Gail. Samuel tries to contact her but she does not respond. When a girl makes a move on Samuel he declines saying he's not ready to move on yet. When he sees an ultrasound of his soon-to-be-born son, he decides that it is time to take responsibility before it is too late. He sells his Porsche buys a family van and gets back together with Rebecca. He marries her and they have the baby together.

Jay sets Max up with his editor Samantha (Mili Avital). Although they share few interests, they are engaged within two weeks. Still, when Max is confronted with the fact that Sam will be the last woman he will sleep with, he proposes a test. Jay will hit on Sam. If she shows no interest, then Max will be confident enough in her loyalty to go ahead with the marriage. However while Jay doesn't want to, the two realize they are obviously made for each other.
The rich fashion designer Julia is tired of living because she knows she is suffering from a malignant cancer. When the woman leaves for her last holiday in Cortina d'Ampezzo, she meets the young and vital Valerio. The two fall in love instantly, but Julia does not reveal her secret to Valerio. When Valerio finds out that she is sick and dying, he decides to pretend to know nothing, continuing his love affair with Julia to the end.
The film begins with an animated introduction featuring a plane heading toward "the Heart of Africa" (more specifically, a region called "Bukuvu"). The plane crashes and George, an infant/toddler, is lost, but enjoys vine swinging and crashing into trees as a running gag. 25 years later, George (Brendan Fraser) has grown into a Tarzan-like man, the "King of the Jungle", who makes his home around Ape mountain. His friends include a sophisticated talking gorilla named Ape (voiced by John Cleese), along with a toco toucan named Tookie, a small capuchin monkey named Little Monkey, and an African elephant named Shep, whom he calls his "dog" (all voiced by Frank Welker).
