In Texas 1858, the Speck brothers, Ace and Dicky, drive black slaves on foot. Among the shackled slaves is Django, sold off and separated from his wife, Broomhilda von Shaft. The Speck brothers are stopped by Dr. King Schultz, a German ex-dentist and bounty hunter from Düsseldorf, Prussia. Schultz asks to buy one of the slaves; when he questions Django about his knowledge of the Brittle brothers, for whom Schultz is carrying a warrant, Ace becomes irritated and aims his shotgun at Schultz. Schultz quickly kills Ace and leaves Dicky at the mercy of the newly freed slaves, who shoot Dicky in the head.
Twenty years after the event in the first Django, the title character has left the violent world of gunslinger to become a monk. Living in a monastery, he wants no more of the violent actions he perpetrated. Later, he learns that along the way, he had a daughter who is working for ruthless criminals in a mine, for which they hope to get rich from the spoils. Determined to find his daughter and nail the bad guys, Django gets some arms and goes on the warpath.
Dance-hall girl Rosie Velez (Divine), lost in the desert, is helped to safety by gunman Abel Wood (Tab Hunter). In the town of Chili Verde, at the saloon of Marguerita Ventura (Lainie Kazan), word of a treasure in gold brings Abel into conflict with outlaw Hard Case Williams (Geoffrey Lewis) and his gang.
One day Sam Clayton (Jack Palance) and his gang arrive in the small town of Juno City where Father John (Lee van Cleef) is the priest of the town church. After having a little bit of fun that involves raping a woman and knifing a man in the back, the gang leaves town, only to be caught by the fearless but unarmed Father John. That night the murderer is broken out of gaol. Vowing revenge, the gang guns him down on the steps of his church and then set about taking control of the town while waiting for the stagecoach. However, little Johnny O'Hara (Leif Garrett) manages to escape with a couple of their horses and rides off to Mexico in the hope of finding the priest's gunfighter brother (also played by Lee van Cleef). They soon meet and set off back across the border to clean up the town. Meanwhile Clayton discovers that he is Johnny's father. Also, some fifteen years prior during the Civil War, Jenny O'Hara (Sybil Danning) had been one of Clayton's victims, adding to the mystique of the situation — as well as to the question of little Johnny's paternity — and now, enhancing the plot, Clayton takes to the idea of being a father.
After the U.S. Civil War, Keoma (Nero), part Indian and part white, returns to his home town to find his half-brothers in alliance with a petty tyrant named Caldwell. Caldwell and his gang rule over the town with an iron fist. With the help of his father and George, an old Black friend, he vows revenge. Keoma also shows compassion when he saves a pregnant woman from a group sent by Caldwell's group to be quarantined in a mine camp full of plague victims. Keoma is constantly visited by the apparition of an older woman ("The Witch") who saved him during the massacre of an Indian camp.

Ho must free his uncle's murderer, Dakota (frequent Spaghetti Western star Lee Van Cleef), the only man who knows Wang's final resting place. Upon recovering the body, the pair discovers clues pointing to buried treasure. Notes on the dead man's body promise that the location of the treasure will be illuminated by a map, divided for security into four segments, each one tattooed on the buttocks of one of the uncle's four mistresses. Armed with photographs of the women, Ho and Dakota form an uneasy alliance and set out in search of the map tattoos and the promised riches.
Jack Beauregard (Henry Fonda) is an aging legendary gunslinger who wants to retire in peace in Europe to get away from young gunmen constantly trying to test themselves against the master. The film opens with three gunmen attempting to ambush Beauregard in a barbershop. After Beauregard has dispatched them, the barber's son asks his father if there is anyone in the world faster than Beauregard, to which the barber replies "Faster than him? Nobody!"
