A Miami businessman, John Stone, receives a parcel from England containing two old bottles of Slivovitz brandy from his recently deceased ancestor, and after drinking both bottles, becomes a vampire. Stone uses his newfound vampire powers to keep his wife, Helena, in a trance as he travels to England to kill the descendents of Abraham Van Helsing that murdered Count Dracula. Meanwhile, Helsing's distant relative, Howard Helsing, pursues Stone with the intent to put the reborn vampire to rest for good.

The Joker and the Penguin break out of Arkham Asylum and race each other to find a stash of stolen money hidden within a crypt in Gotham Cemetery after an inmate, who had hidden it, told them about it. The Joker is quickly intercepted by the Batman and is electrocuted when he falls into a river by his super-charged joy buzzers while the Penguin takes the opportunity to enter the cemetery himself. While searching through a crypt, the Penguin accidentally cuts his hand while using his umbrella-sword to open a coffin he hopes contains the money, finding a body instead. Blood from his hand drips on the corpse's heart, which brings it to life; it is none other than the vampire lord Count Dracula, his body having been moved from Transylvania to Gotham City after his "death." After attacking and turning a watchman at the cemetery into a vampire, Dracula hypnotizes the Penguin into becoming his non-vampiric servant so that he may lead him through Gotham to feed and guard his coffin during the day while he regains his strength.

In 1780, Prince Mamuwalde (William H. Marshall), the ruler of the Abani African nation, seeks the help of Count Dracula (Charles Macaulay) in suppressing the slave trade. Dracula refuses to help, and transforms Mamuwalde into a vampire, whom he names Blacula and imprisons in a sealed coffin. Mamuwalde's wife, Luva (Vonetta McGee), is also imprisoned and dies in captivity. In 1972, the coffin has been purchased as part of an estate by two interior decorators, Bobby McCoy (Ted Harris) and Billy Schaffer (Rick Metzler) and shipped to Los Angeles. Bobby and Billy open the coffin and become Prince Mamuwalde's first victims. At the funeral home where Bobby McCoy's body is laid, Mamuwalde spies on mourning friends Tina Williams (Vonetta McGee), her sister Michelle (Denise Nicholas), and Michelle's boyfriend, Dr. Gordon Thomas (Thalmus Rasulala), a pathologist for the Los Angeles Police Department. Mamuwalde believes Tina is the reincarnation of his deceased wife, Luva. On close investigation of the corpse at the funeral home, Dr. Thomas notices oddities with Bobby McCoy's death that he later concludes to be consistent with vampire folklore.

A group of vampires look for the hidden resting place of Drake, also known as Dracula—the genetic "father" of their species. Count Dracula (Alexander D'Arcy) and his vampire wife (Paula Raymond), hiding behind the pseudonyms of Count and Countess Townsend, lure girls to their castle in the Arizona desert to be drained of blood by their butler George (John Carradine), who then mixes real bloody marys for the couple. Then the real owners of the castle show up, along with Johnny, who is a serial killer or a werewolf depending on which version you watch. The owners refuse to sell, so Dracula wants to force them to sell. In a final confrontation, the vampires are forced to stand in the sunlight and dissolve.

Lawrence Talbot (Lon Chaney, Jr.) is making an urgent call from London to a Florida railway station where Chick Young (Bud Abbott) and Wilbur Grey (Lou Costello) work as baggage clerks. Wilbur answers the phone and Talbot tries to impart to him the danger of a shipment due to arrive for the "McDougal House Of Horrors" (a local wax museum) which purportedly contains the actual bodies of Count Dracula (Béla Lugosi) and the Frankenstein Monster (Glenn Strange). However, before he is able to warn Wilbur, a full moon rises and Talbot transforms into a werewolf, who proceeds to destroy his apartment while Wilbur is on the line. Wilbur, thinking the call is just a prank, hangs up and continues on with his work day. Immediately thereafter, the actual Mr. McDougal (Frank Ferguson) shows up to claim the shipments and, fearing them damaged when Wilbur and Chick mishandle them, demands that the crates be delivered in person so his insurance agent can inspect them.

Renfield is a solicitor traveling to Count Dracula's castle in Transylvania on a business matter. The people in the local village fear that vampires inhabit the castle and warn Renfield not to go there. Renfield refuses to stay at the inn and asks his carriage driver to take him to the Borgo Pass. Renfield is driven to the castle by Dracula's coach, with Dracula disguised as the driver. En route, Renfield sticks his head out the window to ask the driver to slow down, but sees the driver has disappeared; a bat leads the horses.
