A short time after the first film, Mavis (Selena Gomez) and her fiance Johnny (Andy Samberg) are married, with the approval of her father Dracula (Adam Sandler) and the world has become aware of (and unfased by) the existence of monsters. Mavis later reveals to Drac that she is pregnant and a year later, she gives birth to a son named Dennis (Asher Blinkoff), who befriends Wayne's werewolf pup daughter Winnie (Sadie Sandler). Nearing his fifth birthday, Dennis has yet to grow his fangs and Drac worries that his grandson might not gain vampire powers. Noticing the dangers of Transylvania, Mavis starts to consider raising Dennis where Johnny grew up, much to Drac's disapproval.

In the Middle Ages, Vlad Tepes (Luke Evans) is the Prince of Wallachia and Transylvania. As a child, he was a princely hostage of the Ottoman Empire and was trained to be a soldier in the Sultan's elite Janissary corps, where he became their most feared warrior. He was called Vlad the Impaler after slaughtering thousands by impaling them on spears. Eventually sickened by his acts, he put aside his past and returned to rule his domains in peace. One day in the forest, Vlad and his soldiers discover a helmet in a stream. Fearing that an Ottoman scouting party is preparing the way for invasion, they follow the stream to a high mountain cave called Broken Tooth Mountain. Upon entering the cave, they discover the ground of the cave is carpeted in crushed bone; and they are attacked in the dark by an unknown creature (Charles Dance) with shining red eyes, as well as inhuman speed and strength. While his men are killed, Vlad cuts the creature of the cave with his sword before being thrown to the ground. The blood on the sword he used to cut the creature dissolves when exposed to sunlight, and the creature does not pursue Vlad into the sunlight at the mouth of the cave. Returning to his castle, Vlad learns from a local monk that the creature is a vampire, who was once a man who summoned a demon from the depths of hell and made a pact with it for dark powers before being tricked by the demon and cursed to remain in the cave forever until he is released by someone who drinks his blood, then drinks the blood of a human. The person is welcome to share his power in exchange for the freedom to escape the cave.

During the Walpurgis Night in the woods adjacent to the village of Passo Borgo, located at the foot of the Carpathian mountains, a couple of young lovers, Tania and Milos, secretly meet. On her way home, Tania is chased and overcome by a "dark shadow" that kills her. Some time later, Jonathan Harker, a young librarian, arrives at the village hired by Count Dracula, a nobleman from the area. Tania's body mysteriously disappears from the cemetery. In the meantime Harker, before going to Count Dracula's castle, takes the opportunity to visit Lucy Kisslinger, his wife Mina's best friend as well as the daughter of the local mayor.

Flynn Carsen (Noah Wyle) travels to an auction house in England in order to bid on a priceless Ming vase on behalf of the Library. The film takes place in an unidentified city (presumably New York City) in the modern day, and follows the characters of Rufus King (Thomas Downey) and Jacob Van Helsing (Rhett Giles), both of whom have been observing recent attacks made upon young teenagers in the city at night. Van Helsing correctly identifies that the attacks are being made by a group of vampires residing in the city. The vampires are led by a foreign seductress named Countess Bathorly (Christina Rosenberg), who hopes to use the humans to feed her growing vampire clan and to eventually seize control of the city, while at the same time using her growing power to gain the powers of "the Master". Discovering Bathorly's plan, Van Helsing and King begin to hunt down and destroy the vampires one by one, until they finally face the Countess herself and try to kill her once and for all, before her evil consumes the city and allows Dracula's curse to consume the human race.

The Joker and the Penguin break out of Arkham Asylum and race each other to find a stash of stolen money hidden within a crypt in Gotham Cemetery after an inmate, who had hidden it, told them about it. The Joker is quickly intercepted by the Batman and is electrocuted when he falls into a river by his super-charged joy buzzers while the Penguin takes the opportunity to enter the cemetery himself. While searching through a crypt, the Penguin accidentally cuts his hand while using his umbrella-sword to open a coffin he hopes contains the money, finding a body instead. Blood from his hand drips on the corpse's heart, which brings it to life; it is none other than the vampire lord Count Dracula, his body having been moved from Transylvania to Gotham City after his "death." After attacking and turning a watchman at the cemetery into a vampire, Dracula hypnotizes the Penguin into becoming his non-vampiric servant so that he may lead him through Gotham to feed and guard his coffin during the day while he regains his strength.
