Vedam revolves around 5 principal characters. Vivek Chakravarthy (Manoj Manchu) is an upcoming rock star in Bengaluru who hails from a family of decorated army officers. His mother wants him to follow the same path but he isn't interested. He is about to give his first live concert on New Year's Eve with his band-mates in Hyderabad. However, Vivek is late to catch the flight and so the band is forced to travel by road. After sometime, one of their tyres get deflated. Not having a spare, the other friends go to get the tyre inflated leaving Vivek and Lasya (Lekha Washington) alone. Meanwhile, they are attacked by a Hindu racist group. However, they manage to get out of the mess with the help of a Sikh truck driver (who incidentally gets involved in an earlier altercation with Vivek). The band successfully reaches Hyderabad. However they suffer a minor accident there which involves a mother in labour. So Vivek and Lasya take her to a hospital nearby.

Paro Dev (Abhay Deol), the son of a rich businessman, is an insecure narcissist. He and Paro (Mahi Gill) are childhood sweethearts. While separated by distance, Paro and Dev's youthful love only blossoms more. After finishing his studies, Dev returns to Chandigarh and meets Paro. Their endeavor to make love makes for some dark comic moments. When Dev hears rumours about Paro, he immediately believes them and ditches her. The seeds of suspicion are sown here, which the couple are never able to weed out. What makes them fall apart is mutual suspicion and an essentially male vision of how a woman should conduct herself sexually. Paro turns her back on him when she hears him insult her and agrees to marry a man of her parents' choice. On her wedding day, Dev learns that the rumors were false but his ego doesn't let him accept his mistake, and he lets Paro marry someone else.

Shivani Shivaji Roy, a dedicated and brave police officer with Mumbai Police, chases a Delhi-based kingpin, Karan Rastogi, who runs an organised crime cartel involving child trafficking and drugs. Shivani had saved Pyaari from being sold by her uncle and started taking care of her, and since then Pyaari is like a daughter to her. She takes the task more personally and goes beyond her legal rights and duties to nab Karan and save Pyaari. Karan, aware that Shivani is continually monitoring his cartel's activities, phones her, suggesting she not interrupt his business. Determined to catch him, Shivani uses intelligence inputs to track down his associate in Mumbai, who leads her to Karan's close aide, Wakeel, in Delhi. Karan, as a warning to Shivani, dismembers one of Pyaari's fingers and sends it to Shivani's house wrapped in a gift box, and gets Shivani's husband abused and thrashed.

After receiving an unexpected phone call that she has won a spot to participate on a television game show, she becomes obsessed with regaining the youthful appearance she possesses in an old photograph from her son Harry's (Jared Leto) graduation many years earlier. In order to fit into her old red dress seen in the picture, the favorite one of her deceased husband Seymour, she goes on a crash diet. In order to reach her goal sooner, she goes to a doctor to discuss weight loss. The doctor gives her a prescription for weight-loss amphetamine pills throughout the day and a sedative at night. Harry warns her about amphetamine dependence and risk of life-threatening consequences, but she rebuffs him and insists that the chance to be on television has given her a reason to live. As the months go by, Sara's tolerance for the pills adjust and as a result she is no longer able to feel the same high the pills once gave her. When her invitation has still not arrived, she wrongfully increases her dosage from double to triple and, as a result, begins to suffer from amphetamine psychosis. Soon, her delusions worsen and she is driven to the brink of madness when she suffers a hallucination that she appears on the game show as the principal subject while being attacked by her monstrous, anthropomorphized refrigerator.
