A group of animal activists, consisting of Dr. Nikki Riley (Debbie Gibson), Gia (Arden Cho) and Ben (Patrick Hancock), break into a house and steal dozens of exotic pythons before setting them free into the Florida Everglades. Their actions inadvertently cause the snakes to grow into alarming sizes and threaten the ecosystem, as they kill over 72 alligators in a matter of days. Due to the threat of the invasive species, Park Ranger Terry O’Hara (Tiffany) issues permits to the local hunters to exterminate the pythons – much to Nikki’s dismay. However, the hunters themselves – including Terry’s fiancé Justin (Carey Van Dyke) – are quickly eliminated by the giant snakes. Vowing to avenge her fiancé’s death, Terry and Angie (Kathryn Joosten) take boxes of anabolic steroids from Angie’s grandson Manny (Chris Neville) and inject them into dead chickens before feeding them to alligators. Among the steroids is an experimental serum that neutralizes muscle growth inhibitors with a side effect of increased aggression. Meanwhile, Nikki and her group set up cameras all over the Everglades to monitor Terry’s activities. They catch and record Terry and Angie feeding the steroid-laced chickens to the alligators.

A genetics laboratory run by Dr. Irwin Burns (Joseph Ruskin) to research aggressive behavior has an accident, resulting in a chemical fire and explosion, and the escape of a thirty foot long hybrid snake with traits of both the Asian King Cobra (in the film, it is referred to as the African King Cobra, even though the King Cobra species doesn't inhabit the African Subcontinent) and the Eastern Diamondback Rattlesnake. Loose in the countryside and filled with the experimental aggressiveness drug, the snake, nicknamed "Seth", eventually outgrows his woodland prey, it begins hunting the residents of the small brewery town of Filmore.

Paleontologist Robert Trenton (Dean Cain) is called to Northeastern Antarctica near the Indian Ocean to help the FBI build an underground maximum-security military base and prison for the world's most dangerous criminals and terrorists. The prison is dubbed "New Alcatraz" by the staff. While building the prison, the staff accidentally awaken and unleash a prehistoric Boa Constrictor from its 200 million year hibernation. The snake escapes unnoticed until the staff notice a massive hole chewed in the five-foot thick concrete walls. Soon, the staff, and prisoners begin disappearing as they are brutally hunted and eaten by the snake. The FBI agents, Trenton and the rest of the prisoners try to escape, but are hampered as they don't have many weapons. They begin to give up hope as most of the survivors die. However, Trenton manages to kill the snake by kicking it off the helicopter, which it had bitten into, and it is assumed the fall kills the snake.

Suddenly Soriya turned to the snake and Vaha dropped dead by a heart attack. The new wife took everything and blamed Mek as the servant.

In Romania, a female anaconda and a male anaconda captured, are being held in a genetic research facility, Wexel Hall for experimentation. The research project is led by Dr. Amanda Hayes (Crystal Allen) and funded by J.D. Murdoch (John Rhys-Davies), a well-known industrialist. While visiting the facility, Murdoch agitates the male anaconda with a large flashlight, and has it gassed to calm it down. However, as Murdoch, his assistant Pinkus (Ryan McCluskey) and a scientist leave, the anaconda breaks through his enclosure's wall. As it escapes, it kills many of the people working in the lab, though Cupboard escapes the destruction. In looking through the facility, Amanda and another researcher realize that the second (Queen) anaconda has also escaped. Murdoch calls in a team of animal hunters, led by Hammett (David Hasselhoff), to capture both snakes. Amanda and Pinkus go with them. At a small farm in the middle of the woods, the owner is eaten alive by one of the snakes.

J.D. Murdoch, a billionaire suffering from bone cancer, sends his assassin Eugene and his team of henchmen mercenaries to find Peter and the serum of Peter created it that can cure him. He also tells them to be careful of Dr. Amanda Hayes, and to kill her if needed. As Dr. Amanda Hayes, the sole survivor of the previous snake attack, and two officers also go in search of Peter, determined to destroy the serum and kill the snake.

According to a myth, when snakes are of certain age, they can assume human form. Thus, the film begins with a male and a female in their human forms, singing and dancing amorously with each other. When the male turns back into a snake, he is shot by a member of a hunting party, who thought the snake was going to attack the woman. The rest of the story deals with the female taking revenge on the group of friends responsible for her lover's death. In the end, Vijay is the only one alive and the nagin dies before killing him and realizes that she was wrong and that her revenge destroyed many lives just like her own life was destroyed. Near death, she sees her beloved in the sky calling out to her. She dies and reunites with him in heaven.
