Over the two years since the death of Deacon Frost, Blade (Wesley Snipes), the Daywalker, a vampire unaffected by sunlight and who chooses to defend humans and fight other vampires, has been trying to find his mentor Whistler (Kris Kristofferson), who survived his suicide attempt in the first crisis. Blade has been sweeping across Russia and eastern Europe, enlisting the aid of a young man, Scud (Norman Reedus), to design him a new line of gadgetry and weaponry. Blade carves his way through a large gang of vampires, sparing one, Rush (Santiago Segura), when he promises to reveal Whistler's location. Blade tells Rush he will be back for him. Blade finds Whistler locked in a tank by a gang of vampires who were keeping him alive to torture. Blade rescues Whistler and brings him back to the lair, ridding the latter's vampirism with a cure developed by Dr. Karen Jenson.

A group of vampires look for the hidden resting place of Drake, also known as Dracula—the genetic "father" of their species. They find him in a Syrian ziggurat tomb, although not before, in a moment of blood lust, he kills one of them. The vampires capitalize on Blade's reputation as a serial killer and frame him for the killing of a familiar. During the ensuing manhunt, the FBI locate Blade's latest hideout, and, during the siege, Abraham Whistler dies. With his mentor gone and surrounded by human authorities, Blade allows himself to be captured and arrested.

In 1936, a German professor, Richard Wirth, is hosted by the Wollners, a family of German emigrants in West Virginia. Billions of people are killed in World War III, and most of the surviving males are rendered sterile. As a result, humanity resorts to cloning as its primary form of reproduction. Alex McNetti (Rich George) has a rare disease which gives him very brittle bones. His friend Thomas Granger (Brian Paulin) believes that he has found the cure by feeding his friend dead flesh to counteract the terrible disease. However the process does have its side effects including making Alex cough up and vomit worms, and also turning him into a zombie. The dead start to rise from their graves and butcher every living human they see to pieces, including a heavily armed SWAT team. In 1957, The "Sumatran Rat-Monkey" is a hybrid creature that, "according to legend", resulted from the rape of tree monkeys on Skull Island by plague-carrying rats. Stewart McAlden (Bill Ralston), an explorer returning from the depths of the island with his team, is carrying a rat-monkey in a cage and is stopped by fierce warrior natives that demand the return of the monkey. They escape to a waiting jeep, but Stewart gets bitten by the Rat-Monkey. Seeing the mark of the monkey's bite on his right hand, Stewart's men hold down the explorer and amputate it. A bite mark is then seen on his left arm, which results in the removal of that limb. Finally, they see a set of bloody scratches on Stewart's forehead and kill him. The captured rat-monkey is shipped to Wellington Zoo in New Zealand.

When their originally planned outing is cancelled, four friends go on a hunting trip in Texas.
