American Splendor retrace la véritable histoire du scénariste de comics Harvey Pekar. Au début du film, celui-ci mène une existence plutôt morose entre son travail de documentaliste, quelques conversations sur tout et rien avec ses collègues et quelques connaissances, et sa passion pour le jazz, qu'il assouvit en recherchant des pièces rares dans les garage sales (équivalent américain des vide-greniers) ou les magasins d'occasion. C'est ainsi qu'il fait la connaissance du célèbre dessinateur de bande dessinée Robert Crumb, alors inconnu, qui est lui aussi collectionneur. Plusieurs années plus tard, alors que Crumb est devenu célèbre, Harvey lui soumet les story-boards d'un projet de bande dessinée basée sur sa vie, lui proposant d'en assurer le dessin. Crumb accepte, et contre toute attente, cette série intitulée American Splendor relatant une vie simple, banale, obtient un grand succès. Harvey Pekar acquiert ainsi une certaine célébrité aux États-Unis, dont les conséquences sur sa vie quotidienne sont présentées dans le film. En particulier, American Splendor lui permet de rencontrer sa future femme Joyce Brabner, qui écrit elle aussi des comics, et avec qui il va réaliser d'autres histoires autobiographiques, également intégrées en partie dans le film (notamment Our Cancer Year).

Riggan Thomson (Michael Keaton) is a faded Hollywood actor best known for playing the superhero Birdman decades ago in a series of films. He is often tormented by the mocking, critical inner voice of Birdman, and frequently visualizes himself performing feats of levitation and telekinesis. The film opens with a quotation from Raymond Carver stating that the main fulfillment in life is related to having once been loved, then cuts to a dramatic atmospheric disturbance resembling a meteor descending in flames. The film then cuts to a surreal shot depicting Riggan as levitating in his stage room. Riggan hopes to reinvent his career by writing, directing, and starring in a Broadway production of a loosely based adaptation of Carver's short story "What We Talk About When We Talk About Love".

Woodrow "Woody" Wilkins is an imaginative, yet eccentric, comic book writer and illustrator who demands a sense of realism for his comic book hero "Condorman", to the point where he crafts a Condorman flying suit of his own and launches himself off the Eiffel Tower. The test flight fails as his right wing breaks, sending him crashing into the Seine River. Later after the incident, Woody is asked by his friend, CIA file clerk Harry, to perform what appears to be a civilian paper swap in Istanbul. Upon arriving in Istanbul, he meets a beautiful Soviet named Natalia Rambova, who poses as the Soviet civilian with whom the exchange is supposed to take place, but it is later revealed that she is in fact a KGB spy. Woody does not tell Natalia his real name, and instead fabricates his identity to her as a top American agent code-named "Condorman". During the encounter, Woody fends off a group of would-be assassins and saves her life by sheer luck before accomplishing the paper trade. Impressed by Woody, and disgusted by how she was treated by her lover/boss Krokov when she returns to Moscow, Natalia decides to defect and asks the CIA to have "Condorman" be the agent that helps her.

In 1945 Las Vegas, World War II veteran Frank Harris (Brad Pitt) returns to his mother. Whiskers finds Frank useful enough to run things in the Cool World while he is gone to the real world.

Joey Wellman, an American cartoonist from Cleveland now largely forgotten at home, visits France with his partner Lena to attend an exhibition in Paris about the comic strip (bande dessinée) which features his work. Elsie is naively infatuated with French literature, and is trying to secure an introduction to the brilliant university professor Christian Gauthier, an expert on Flaubert but also an enthusiast for comic books. The meeting of father and daughter goes badly, but Elsie is persuaded to join Joey and Lena for the weekend at the country house of Gauthier's mother, Isabelle. During a comic-themed masquerade party, all of the characters are made to reconsider their present and past relationships.

American caricaturist Jim Crocker is a popular womanizer in London known by his pen name 'Piccadilly Jim'.
