Having successfully completed his final mission three years prior, which was to retrieve a truck load of plutonium stolen from a US military base in Croatia by freelance international terrorist Stavros, government anti-terrorist agent Jack Paul Quinn is relaxing by his pool in Southern France with Kathryn, his pregnant wife. Quinn is approached by a government representative who tells him that Stavros, Quinn's nemesis, has become active again and tries to convince Quinn to come out of retirement telling Quinn that he ‘can’t retire until he [Stavros] does’. Quinn is reluctant to return to duty but agrees after the same representative is killed by Stavros shortly after the meeting with Quinn.
The bulk of the story takes place in Monaco. Charming attractive Alex (Romain Duris), his sister Mélanie (Julie Ferrier), and her husband Marc (François Damiens), operate a unique business for concerned third-party clients—breaking up relationships, but only where the woman is "not knowingly unhappy." The trio concoct elaborate, custom, and sometimes expensive ruses to deceive the women. After each woman has fallen for his act, Alex tells her she has made him come alive again, but that it is too late for him. The women presumably leave their relationships to seek men who make them feel the way Alex has.
In Marseille, France, a young Vespa delivery boy named Daniel (Samy Naceri) has just finished his last delivery for a local pizza parlor, Pizza Joe, owned by Joe (Georges Neri), his boss, also beating his old record, as he is planning to quit the job to pursue a career as a taxi driver. As he reunites with his girlfriend Lilly (Marion Cotillard), a large group of delivery boys throw a huge celebration in honor of his departure as he goes with Lilly to his apartment.
Frank Martin (Jason Statham) is a highly skilled driver known only as "The Transporter." He will transport anything, no questions asked, always on time, and he is known as the best in the business. He strictly follows three rules when transporting: Rule Number 1: "Never change the deal", Rule Number 2: "No names", and Rule Number 3: "Never open the package." Frank has been hired to transport some robbers from a bank heist. On delivery to their destination, they foist an extra man in the car so he refuses to drive until they decide to kill one robber. Later they offer new money on Frank to drive further from the city. He refuses the deal. The robbers escape in another car, and Frank leaves.
In the prologue, a young Max Skinner, whose parents have died in an accident, spends his childhood summer holidays learning to appreciate the finer things in life at his Uncle Henry's vineyard estate in Provence in south-eastern France. Some 25 years later, Max is an unethical yet very successful hard-working London-based bond trader with a large portion sense of humour.
Lawrence Jamieson is an intelligent and sophisticated British con artist operating in the French Riviera with the help of manservant Arthur and corrupt police official Andre. He seduces wealthy and morally suspect women and steals their money. His only concern is a con artist known as "The Jackal" who has been preying on other wealthy victims.
In Marseille, an undercover detective is following Alain Charnier (Fernando Rey), a wealthy French criminal who runs the largest heroin-smuggling syndicate in the world. The policeman is assassinated by Charnier's hitman, Pierre Nicoli (Marcel Bozzuffi). Charnier plans to smuggle $32 million worth of heroin into the United States by hiding it in the car of his unsuspecting friend, French television personality Henri Devereaux (Frédéric de Pasquale).

Sarah's publisher, John Bosload, offers her his country house near Lacoste, France for some rest and relaxation. After becoming comfortable with the run of the house, Sarah's quietude is disrupted by a young woman claiming to be the publisher's daughter, Julie. She shows up one night claiming to be taking time off from work herself. She also claims that her mother used to be Bosload's mistress, but that he would not leave his family.
