In 2009, a genetically re-engineered measles virus, originally created as a cure for cancer, turns into a lethal strain which kills 90% of those it infects, and mutates the remaining 10% into predatory, nocturnal mutants called "Darkseekers" who are extremely vulnerable to sunlight and other sources of UV. Three years after the outbreak, US Army virologist Lieutenant Colonel Robert Neville (Will Smith) lives an isolated life in the ruins of New York City, which is now deserted, unsure if any other uninfected humans are left in the world.
A zombie outbreak erupts in metropolitan areas around the world; those bitten by the creatures become zombies themselves within twelve seconds. Retired UN investigator Gerry Lane, his wife Karen, and their two daughters manage to escape the outbreak in Philadelphia and take shelter with a family in Newark while waiting for special evacuation the next day, thanks to Gerry's friend UN Deputy Secretary-General Thierry Umuntoni. They find a shelter for the night while waiting for a rescue helicopter. Their hosts choose not to accompany them when they flee the next day, and are subsequently bitten and changed by zombies. Their son, Tommy, manages to escape, and Gerry's family takes him in. They are flown to an offshore U.S. Navy carrier group near New York where Umuntoni is overseeing the remaining worldwide governments' reactions.

Mavis later reveals to Drac that she is pregnant and a year later, she gives birth to a son named Dennis (Asher Blinkoff), who befriends Wayne's werewolf pup daughter Winnie (Sadie Sandler). Nearing his fifth birthday, Dennis has yet to grow his fangs and Drac worries that his grandson might not gain vampire powers. Noticing the dangers of Transylvania, Mavis starts to consider raising Dennis where Johnny grew up, much to Drac's disapproval.

The film begins with a young boy, Thomas, taken in by Ava Paige (Patricia Clarkson) and troops from the organization WCKD, alongside other children, after being dropped off by his mother for his own safety. Years later, shortly after being rescued from the maze, Thomas (Dylan O'Brien) and the remaining Gladers - Newt (Thomas Brodie-Sangster), Minho (Ki Hong Lee), Teresa Agnes (Kaya Scodelario), Frypan (Dexter Darden) and Winston (Alexander Flores) - are taken to a facility run by Mr. Janson (Aidan Gillen). He explains that the facility is a safe haven, protecting them from WCKD, the Flare virus and the Cranks, and provides them clothes, food, and sanitation, where there are other survivors of multiple mazes are also present. Thomas becomes curious about their activities and manages to find out what is really happening behind the closed doors. With help from Aris Jones (Jacob Lofland), the first maze survivor to end up in the facility, it is revealed that WCKD's leader, Ava, is still alive and that Mr. Janson is working for WCKD, as they discuss about the experiment on the Immunes and that Janson has yet to find The Right Arm, a resistance group located at the mountains. The group, including Aris, escapes the facility, as Janson orders his WCKD troops to track them down, where they find themselves in the deserted outside world - the Scorch.
Picking up immediately after the events in Resident Evil: Retribution, humanity is on its last legs after Alice is betrayed by Wesker in Washington D.C. As the only survivor of what was meant to be humanity's final stand against the undead hordes, Alice returns to where the nightmare began, Raccoon City, where the Umbrella Corporation is gathering its forces for a final strike against the only remaining survivors of the apocalypse. In a race against time, Alice joins forces with old friends, and an unlikely ally, in a battle with undead hordes and new mutant monsters. Between losing her superhuman abilities and Umbrella's impending attack, this will be Alice's most difficult adventure as she fights to save humanity.
A year after the events of Extinction, the Alice clones (Milla Jovovich) attempt to kill Albert Wesker (Shawn Roberts) at the current Umbrella Headquarters, Tokyo. The clones manage to kill the entire branch, but Wesker makes his way to his private charter tiltrotor plane and escapes before activating a bomb, leaving a massive sinkhole. However, the real Alice boarded his aircraft beforehand. As she pulls a gun on him, Wesker injects her with Anti-Virus before the lack of a pilot makes the aircraft crash in the mountains outside Tokyo. Only Alice emerges from the crash alive.
Alice (Milla Jovovich) and the others on the Umbrella Corporation freighter Arcadia face an attack by a fleet of tiltrotors led by Alice's former ally, Jill Valentine (Sienna Guillory), who is being mind controlled by Umbrella through a scarab device attached to her chest. Alice causes a tiltrotor to crash into Arcadia, resulting in an explosion that knocks her out and into the water. The fates of Chris Redfield, Claire Redfield and K-Mart are ambiguous.
Alice (Milla Jovovich) wakes up disoriented in a mansion. She wanders through the halls, where she defeats several obstacles, including a laser beam obstacle (from the first film) and a giant blade that falls from the ceiling. However, she is eventually killed by a miniature machine gun. Her body is dumped into a pit filled with hundreds of Alice clones. The camera zooms out to show a shack camouflaging the facility's above-ground entrance - with the rest of the facility located underground, surrounded by a high fence and thousands of zombies.
30 days after the contamination of The Hive seen in Resident Evil (2002), the Umbrella Corporation unwisely sends in a research team to re-open the complex and investigate the incident, since no one survived except Alice and Matt Addison, and as Alice was experimented on, Matt was put into a mysterious "Nemesis Program". When the team reprograms and opens the sealed blast doors, they are slaughtered by the massive crowd of infected.

In the small town of Blithe Hollow, Massachusetts, Norman Babcock (Kodi Smit-McPhee) is an 11-year-old boy who speaks with the dead, including his late grandmother (Elaine Stritch) and various ghosts in town. Almost no one believes him and he is isolated emotionally from his family while being ridiculed by his peers. His friend, Neil Downe (Tucker Albrizzi), is an overweight boy who is bullied himself and finds in Norman a kindred spirit. During rehearsal of a school play commemorating the town's execution of a witch three centuries ago, Norman has a vision of the town's past in which he is pursued through the woods by townsfolk on a witch hunt. Afterward, the boys are confronted by his estranged and seemingly deranged uncle Mr. Prenderghast (John Goodman) who tells his nephew that he soon must take up his regular ritual to protect the town. Soon after this encounter, Prenderghast dies from a sudden stroke. During the official performance of the school play Norman has another vision, creating a public spectacle of himself which leads to his father (Jeff Garlin) grounding him. His mother (Leslie Mann) tells him that his father's stern manner is because he is afraid for him. The next day, Norman sees Prenderghast's spirit who tells him that the ritual must be performed with a certain book before sundown that day. Norman is at first reluctant to go because he is scared but his grandmother tells him it is all right to be scared as long as he does not let it change who he is. Norman sets off to retrieve the book from Prenderghast's house (having to take it from his corpse).
