In San Francisco in the 1910s, the clownish musical band that runs Firehouse 12 often drowns out alarms with their loud music. Smokey Hinkle (Buddy Hackett), a bumbling inventor and scion of a renowned firefighting family, buys retired firehouse horse Emma to pull his buggy, but when Emma hears an alarm, she follows the fire trucks at top speed. Smokey arrives at the fire at the same time as his pal, fireman Smitty (Hugh O'Brian), who points out that the blaze is in Smokey's own house. Smokey uses his new invention, an ingenious fire extinguisher, to put out the flames, prompting Smitty to hire Smokey as a fireman in exchange for half of the profits from the sale of the extinguisher.

The basic plot remains the same as in the stage play and the 1936 film version. When the Cotton Blossom, Cap'n Andy Hawks's show boat, arrives in a Mississippi town to give a performance, a fistfight breaks out between leading man Steve Baker (Robert Sterling), and Pete (Leif Erickson), the boat's engineer who has been making passes at Steve's wife, leading lady Julie La Verne (Ava Gardner). But Cap'n Andy (Joe E. Brown) pretends to the assembled crowd that the two were really previewing a scene from one of the boat's melodramas. Lou Francis (Lou Costello) and Bud Alexander (Bud Abbott) have just graduated from a private detective school. Tommy Nelson (Arthur Franz), a middleweight boxer, comes to them with their first case. Tommy recently escaped from jail after being accused of murdering his manager, and asks the duo to accompany him on a visit to his fiancée, Helen Gray (Nancy Guild). He wants her uncle, Dr. Philip Gray (Gavin Muir), to inject him with a special serum he has developed which will render Tommy invisible, and hopes to use the newfound invisibility to investigate his manager's murder and prove his innocence. Dr. Gray adamantly refuses, arguing that the serum is still unstable, recalling that the formula's discoverer John Griffin was driven insane by the formula and did not become visible again until after he was killed. However, as the police arrive Tommy injects himself with it and successfully becomes invisible. Detective Roberts (William Frawley) questions Dr. Gray and Helen while Bud and Lou search for Tommy.
