Moore commence sa carrière au Royaume-Uni avant de travailler pour des éditeurs américains. Célèbre pour avoir rendu dans les années 1980 les comics plus matures et plus littéraires, il a également beaucoup apporté à la forme du médium, par des effets de mise en page inédits jusqu'alors. Ses influences, très diverses, comptent des auteurs comme William S. Burroughs, Thomas Pynchon, Robert Mayer et Iain Sinclair, des écrivains de science-fiction comme Michael Moorcock et d'horreur comme H.P. Lovecraft, Clive Barker, des cinéastes comme Nicolas Roeg. En bande dessinée, l'influence de Bryan Talbot, précurseur de la bande dessinée « adulte » avec The Adventures of Luther Arkwright, a été déterminante.
Végétarien et connu pour ses opinions anarchistes, il se présente par ailleurs comme un magicien et un adorateur de Glycon, une divinité-serpent romaine.
Il reprend Marvelman et crée V pour Vendetta dans les magazines Doctor Who Weekly et Warrior. V pour Vendetta connaît un grand succès et obtient de nombreux prix dont celui de meilleur album étranger à Angoulême en 1990. Cela vaut à Alan Moore d'être engagé en 1983 par l'éditeur DC Comics pour travailler sur la série The Saga of the Swamp Thing. Il revisite brillamment ce comic des années 1970 et obtient un succès retentissant.

Alan Moore se brouille avec DC Comics, pour des raisons de droits d'auteurs notamment, en 1988 et monte sa propre maison d'édition Mad Love. Il commence alors From Hell qui obtient le prix de la critique à Angoulême en 2001 et fait l'objet d'une adaptation cinématographique la même année.
En 1991 Alan Moore commence à travailler sur une BD érotique, Lost Girls, participe à un projet multimédia : The Moon and Serpent Grand Egyptian Theatre of Marvels en 1994, publie un roman intitulé Voice of the Fire en 1996.
En 1996, encore, il reprend la série Supreme (un pseudo-Superman, initialement œuvre de Rob Liefeld) en collaboration avec les dessinateurs Joe Bennett, Alex Ross et Rick Veitch, où il continue son travail d'exploration nostalgique de l'époque de l'âge d'or des comics. Il publie aussi, toujours pour Rob Liefeld et le Studio Awesome, Judgement Day où, à l'instar d'un Crisis on Infinite Earths des années 1990, il propose de refonder tout un univers.
En 1999, il sort la collection ABC. Elle comprend entre autres La Ligue des gentlemen extraordinaires adapté au cinéma en 2003, Top Ten, Promethea et Tom Strong.
Celle-ci devait être éditée par la compagnie de Jim Lee, Wildstorm. Celle-ci ayant été vendue entre-temps à DC Comics, Alan Moore accepta plutôt de mauvaise grâce cet accord et a annoncé que l'univers du label se terminerait avec la fin du 32e épisode de Promethea.
Jusqu'ici les films tirés de l'œuvre de Moore ont été des adaptations hollywoodiennes ayant finalement peu à voir avec l'original au point que Moore refusa même de toucher les droits sur les films tirés de ses œuvres. Il accorde peu de crédit au cinéma qui, en raison du nombre d'intervenants qu'il requiert, ne lui semble pas permettre la même liberté d'expression que l'écriture.
En 2006, James McTeigue réalise l'adaptation de V pour Vendetta, sur un scénario des sœurs Wachowski. En 2009, c'est au tour de Watchmen d'être adapté sur grand écran par Zack Snyder, un premier projet d'adaptation par Terry Gilliam ayant été abandonné par le producteur Joel Silver.
Alan Moore a reçu neuf fois le prix du meilleur scénariste aux Eisner Award depuis 1988, trois fois aux Jack Kirby Award, et sept fois aux Harvey Award. Cette liste de récompenses fait probablement de lui le scénariste le plus primé dans le domaine du comic.

Il nait à Northampton dans une famille modeste. Après une scolarité difficile (il est exclu de son lycée pour avoir vendu du LSD), il monte le magazine Embryo avec des amis à l'âge de 18 ans. Il se marie en 1974 et a deux enfants. In the late 2020s the world is in turmoil, with the United States fractured as a result of prolonged conflict and a pandemic of the "St. Mary's Virus" ravaging Europe. The United Kingdom is ruled as a fascist police state by the Norsefire Party. Political opponents, immigrants, Muslims, homosexuals and other "undesirables" are imprisoned in concentration camps. On November 4, a Guy Fawkes-masked vigilante identifing himself as "V" (Hugo Weaving) rescues Evey Hammond (Natalie Portman), an employee of the state-run British Television Network (BTN), from members of the "Fingermen" secret police while she is out past curfew. From a rooftop, they watch his demolition of the Old Bailey criminal court building, accompanied by fireworks and the 1812 Overture. Inspector Finch (Stephen Rea), Scotland Yard's Chief of Police, is tasked with investigating V's activities while BTN declares the incident an "emergency demolition". V interrupts the broadcast to claim responsibility and urges the people of Britain to rise up against their government. He asks them to meet him exactly one year later, on November 5, Guy Fawkes Night, outside the Houses of Parliament, which he promises to destroy. During the broadcast, the police attempt to capture V. Evey helps him escape but is knocked unconscious.

In 1899, men dressed as German soldiers, using a large armored vehicle and wielding machine guns, attacked the Bank of England and stole Leonardo da Vinci’s layouts of Venice’s foundations. Shortly after, led by their leader the “Fantom”, men dressed as British officers, again using highly advanced technology for the time, kidnapped German scientists and destroyed a factory, causing tension between the United Kingdom and Germany, which could lead to an international war. Sanderson Reed (Tom Goodman-Hill) of the British Empire ventures to Kenya, visiting a Gentleman's club to recruit world-renowned hunter and adventurer Allan Quatermain (Sean Connery). Quatermain, retired and mourning the loss of his son after his last adventure, has no interest in serving the British Empire further. After armed men attempt to assassinate Quatermain, he is left with no-other choice but to agree to go with Reed to London and meet with Reed's superior officer to help keep the threat of a war from spilling into Africa.

In 1888, Mary Kelly (Heather Graham) and her small group of London prostitutes trudge through unrelenting daily misery. When their friend Ann Crook (Joanna Page) is kidnapped, they are drawn into a conspiracy with links higher up than they could possibly imagine. The kidnapping is soon followed by the gruesome murder of another woman, Martha Tabram (Samantha Spiro); and it becomes apparent that they are being hunted down, one by one as various prostitutes are murdered and mutilated post-mortem.
